基于重测序的晋汾52及其突变体抗性差异分析 被引量:3

摘要 [目的]名优谷子品种晋谷21号是由晋汾52辐射诱变而来,在病害抗性方面存在明显差异,本文旨在探明晋汾52和及其突变体品种晋谷21对植物病害抗/感的原因。[方法]通过利用全基因组重测序数据,对2个谷子品种在整个基因组水平和NBS类型基因中存在的差异和变异位点进行分析。[结果]对2个谷子品种在整个基因组中SNP、InDel、SNV和SV等位点分析发现,SNP的突变类型以纯合突变为主,InDel和SV突变数目在第8条染色最多;对2个品种的NBS类型基因分析,差异也主要存在于第8条染色体上,其变异类型和变异数目也是最多的。推测晋谷21的较晋汾52更感病很可能与NBS类基因的结构域缺失有关。[结论]2个品种NBS类型基因结构的差异可能是造成抗感差异的原因,该研究结果可为进一步分析确定抗病基因关键差异位点提供数据参考和理论依据。 [Objective]Jingu 21 is a high-quality millet variety originated from Jinfen52 dry seeds by radiation mutation with cobalt60 gamma-ray,and bred by continuous plant selection.However,a significant variation of disease resistance between JinFen52 and Jingu 21 was noticed.The aim of present paper was to explore the causes of the variation between mentioned two varieties using genome resequence.[Methods]Resequencing data of two millet varieties were collected and compared.Differences and loci variation at whole genome level and NBS type gene were analyzed.[Results]Study demonstrated that the main mutation type of SNP was homozygous mutant,and the greatest mutation number of InDel and SV was detected on the eighth chromosome.Analysis of NBS genotype of two varieties displayed the same results that eighth chromosome owned the highest mutation number with the same type and variation number.Therefore,we speculated that the deletion of NBS gene domain might contribute to the disease resistance variation that Jingu 21 was more susceptible than Jinfen 52.[Conclusion]The difference in NBS genes structure would be the reason of resistance variation between two tested varieties.The results can provide data reference and theoretical basis for further analysis of resistant gene locus.
