神香草提取物对哮喘模型大鼠血清代谢物的影响 被引量:3

Effects of extract of Hyssopus officinalis L. on serum metabolites in asthma model rats

摘要 目的探讨神香草提取物(总黄酮含量高于50%以上,SXCF)对哮喘模型大鼠及其血清代谢物变化的影响。方法在建立哮喘模型大鼠的基础上干预SXCF,将60只SD大鼠随机分为正常对照组(生理盐水0.3mg/kg体质量)、哮喘模型组(生理盐水0.3mg/kg体质量)、阳性对照组(醋酸地塞米松0.3mg/kg体质量)、SXCF低、中、高剂量组(SXCF0.2、0.4、0.8g/kg体质量),每组10只,观察各组大鼠肺组织病理改变,测定大鼠血清免疫球蛋白E(IgE)水平,进行模型评价,观察SXCF对哮喘模型大鼠的影响。收集各组大鼠血清,采用核磁共振波谱(1 H-NMR)技术测定大鼠血清,建立大鼠血清代谢指纹图谱,运用偏最小二乘判别式(PLS-DA)法,正交偏最小二乘判别式(OPLS-DA)法分析大鼠血清中代谢物的差异。结果与正常对照组比较,哮喘模型组大鼠支气管壁及血管周围有炎细胞浸润,淋巴细胞及嗜酸性粒细胞,支气管黏膜皱壁增多,有黏膜脱落现象,平滑肌增厚,管腔狭窄痉挛,融合成肺气肿及肺大泡,符合哮喘的病理学改变,大鼠血清总IgE水平升高。与哮喘模型组比较,SXCF组大鼠肺组织病理改变情况减轻,大鼠血清IgE水平下降。哮喘模型组大鼠血清中亮氨酸、丙氨酸、乙酸、谷氨酸、肌酸、甘氨酸、肉碱、α葡萄糖、β葡萄糖含量降低,乳酸、不饱和脂类、丙酮含量升高,差异有统计学意义(P<0.05)。与哮喘模型组比较,SXCF各剂量组大鼠血清中亮氨酸、肌酸、谷氨酸、肉碱、β葡萄糖含量升高,乳酸、丙酮含量降低,差异有统计学意义(P<0.05)。结论 SXCF对哮喘大鼠血清代谢有一定的影响,SXCF能改善哮喘大鼠的一般情况,减轻肺组织病理改变,降低大鼠血清IgE水平,并对哮喘大鼠血清代谢物有影响,从而起到治疗哮喘的作用。其作用机制可能是通过作用于哮喘大鼠体内氨基酸、能量、脂肪和糖代谢途径而发挥预防抗哮喘作用。 Objective To investigate the effect of Hyssopus officinalis L.extract(over 50% total flavonoids content,SXCF)on asthma model rats and their serum metabolites.Methods Establish rat asthma model and intervene with SXCF,total of 60 SD rats were randomly divided into normal control group(based on weight given physiological saline 0.3 mg/kg),asthma model group(based on weight given physiological saline 0.3 mg/kg),positive control group(based on weigh given dexamethasone acetate 0.3 mg/kg),SXCF low,middle and high dose groups(based on weigh given SXCF0.2,0.4,0.8 g/kg).10 SD rats for each group.To observe the effect of SXCF on asthmatic rats,by observing the pathological changes of lung tissue in rats and measuring the Ig-E levels in each group.The serum of rats in each group were determined by nuclear magnetic resonance spectroscopy-hydrogen spectrum(^1 H-NMR)technique to establish the metabolic fingerprint of rat serum.To analyze the metabolites in rat serum by PLS-DA and OPLS-DA method.Results Compared with the normal control group,there were lymphocyte and eosinophils and other inflammatory cell infiltration around the bronchial wall and blood vessels in asthma model rats.Compared with the normal control group,the bronchial mucous membranes in the model group were increased,model group had mucosal shedding and thickened smooth muscles,the bronchial stenosis became emphysema and pulmonary bullae,the Ig-E levels were increased.Compared with the asthma model group,the pathological changes of lung tissue in the positive control group and SXCF different dose groups were reduced,and serum Ig-E levels were decreased.Compared with the normal control group the levels of leucine,alanine,acetic acid,glutamic acid,creatine,glycine,carnitine,alpha glucose,beta glucose in the serum of rats were decreased and the levels of lactic acid,unsaturated lipids and acetone were increased.The results showed a statistically significant difference(P〈0.05).Compared with the asthma model group the levels of leucine,alanine,acetic acid,glutamic acid,creatine,glycine,carnitine and β-glucose in the serum of rats in SXCF different dose groups were increased,and the levels of lactic acid,acetone,unsaturated lipids were decreased.The results showed a statistically significant difference(P〈0.05).Conclusion SXCF can improve the general condition of asthmatic rats and have an effect on the serum metabolites of asthmatic rats by reducing the level of serum Ig-E,the pathological changes of lung tissue,and by acting on amino acid,energy,fat and glucose metabolism pathways in asthmatic rats.
