黄芩素的体外抗炎及抗氧化活性研究 被引量:54

摘要 采用LPS刺激小鼠单核巨噬细胞RAW 264.7释放炎症介质NO、PGE2以及IL-6、TNF-α等炎症因子,以MTT法评价细胞毒性,Griess法检测NO的含量,ELISA法检测PGE2、IL-6和TNF-α水平,Western blot法检测iNOS、COX-2及内参蛋白β-actin表达水平,研究黄芩素的抗炎活性及作用机制.测定黄芩素对DPPH自由基、超氧阴离子的清除活性,评价其抗氧化活性.结果表明,黄芩素能显著抑制炎症介质NO和PGE2的产生以及炎症因子TNF-α和IL-6的释放,明显下调iNOS、COX-2蛋白表达水平,对DPPH自由基、超氧阴离子均具有较强的清除活性,且呈良好剂量依赖关系.通过对黄芩素的抗炎、抗氧化作用的研究,可以为黄芩素及相关制剂的临床应用提供一定的理论依据. In order to underline the anti-inflammatory,antioxidative activity and the molecular mechanism of baicalein,mouse macrophage RAW 264. 7 cells are stimulated by using LPS to release inflammatory mediators such as NO,PGE2,IL-6 and TNF-α. MTT method is used to determine the cytotoxicity,Griess method is used to determine the production of NO,ELISA kits are used to determine the levels of PGE2,TNF-α or IL-6,Western blot analysis is used to detect the protein expression of iNOS,COX-2 and the β-actin. The antioxidative effect of baicalein is evaluated through the determination of scavenging activity on DPPH free radical and superoxide anion. The results show that baicalein can significantly inhibit the overproduction of NO,PGE2,TNF-α,and IL-6,and that it can completely down-regulate the protein expression of iNOS and COX-2. Baicalein shows strong scavenging activity on DPPH free radical and superoxide anion in a dose dependent manner. These studies on the anti-inflammatory and antioxidative effect of baicalein may provide a theoretical basis for the clinical application of baicalein and its related preparations.
