摘要
目的采用高效液相色谱法(HPLC)和紫外分光光度法(UV)分别测定芦丁片中芦丁的含量并进行比较。方法 HPLC法采用色谱柱为Venusil MP C18(5μm,4.6×250 mm),流动相为甲醇-0.2%冰醋酸(40∶60),流速为1.0 ml/min,检测波长为254 nm,柱温为30℃,进样量20μl。紫外分光光度计法以75%的乙醇为溶剂,检测波长为500 nm。结果紫外分光光度法中芦丁浓度在5~100μg/ml时,吸光度和浓度呈良好的线性关系(r=0.9990),平均回收率为99.36%,RSD为1.25%。芦丁浓度在20~100μg/ml时,高相液相色谱法测得线性关系良好(r=0.9996),平均回收率为100.3%,RSD为1.58%。结论采用F检验对HPLC法和UV法的芦丁含量测定结果进行检查,结果显示两者测得结果没有显著性差异,两种方法均可用于芦丁片中芦丁的含量测定。
Objective: To determine the content of Rutin in Rutinum Tablets by high performance liquid chromatography( HPLC) and ultraviolet spectrophotometry( UV). Methods: The HPLC method with the column was Venusil MP C18( 5μm,4. 6 × 250 mm),the mobile phase was methanol-0. 2% acetic acid( 40∶ 60),the flow rate was 1. 0 ml/min,the detection wavelength was 254 nm,the column temperature is 30℃,sample volume 20μl. UV spectrophotometer was used with 75% ethanol as solvent,and the detection wavelength was 500 nm. Results: The UV Spectrophotometry in the concentration of Rutin in 5-100 g/ml,a good linear relationship between the absorbance and the concentration( r = 0. 9990),the average recovery rate was 99. 36%,RSD was 1. 25%. The concentration of Rutin in 20 g/ml - 100 g/ml,high performance liquid chromatography measured a good linear relationship( r = 0. 9996),the average recovery rate was 100. 3%,RSD was 1. 58%. Conclusion: The results of rutin content determination by HPLC method and UV method are checked by F test. The results show that there is no significant difference between the two methods,and the two methods can be used for the determination of Rutin in Rutinum Tablets.
