摘要
为了快速高效地鉴定杂交种纯度,建立完善的西瓜杂交种纯度SSR分子标记鉴定技术体系。利用30对杂合率高、多态性高的SSR引物对‘豫艺360’‘豫艺黑小宝’‘国豫二号’和‘甜王’这4份在河南省广泛种植的西瓜杂交种父母本进行筛选,筛选出7对条带清晰、多态性较好的引物。通过PCR反应体系扩增及凝胶电泳进行纯度分析,对4份杂交种的F_1样本进行鉴定,纯度分别为91.1%、93.2%、92.6%、91.4%,将分子标记鉴定结果与田间形态学鉴定的结果相比对,两者偏差≤2%,在允许范围之内,表现出高度一致。
SSR markers are specific DNA fragments which can detect certain differences between individuals or populations at the DNA level. In order to identify quickly and efficiently the purity of hybrids, a perfect SSR molecular marker identification technology system for watermelon hybrids was established. In this study, 30 pairs of SSR primers were used to screen the polymorphism of‘Yuyi 360',‘Yuyi Heixiaobao',‘Guoyu 2'and‘Tianwang'4 which were the representative watermelon hybrid parents, 7 of these primers were selected with clear bands and good polymorphism according to the test. The PCR reaction system was amplified and the gel electrophoresis was used for purity analysis. 550 F1 samples of four hybrids were used to verify the accuracy of the markers, and the purity was 91.1%, 93.2%, 92.6% and91.4%, respectively. The results were identified by molecular markers and field morphology. The results showed a high degree of agreement and the errors were within the allowable range of ≤2%. Therefore, SSR molecular marker can be used to replace the identification of traditional field morphological characters and identify rapidly the purity of watermelon hybrids.
