ω-3多不饱和脂肪酸对梗阻性黄疸大鼠炎症反应和肝功能的影响 被引量:3

摘要 目的:探讨ω-3多不饱和脂肪酸对梗阻性黄疸大鼠炎症反应和肝功能的影响。方法:将72只成年雌性Wisar大鼠随机分成4组(n=18):OJ+ω-3组、OJ+NS组、SO+ω-3组、SO+NS组。OJ+ω-3组、OJ+NS组采用双重结扎胆总管方法构建梗阻性黄疸大鼠模型;SO+ω-3组、SO+NS组只行开腹手术。OJ+ω-3组、SO+ω-3组给予ω-3鱼油脂肪乳尾静脉注射[5 mL/(kg·d),1次/d],OJ+NS组与SO+NS组大鼠给予同等体积生理盐水尾静脉注射。建模成功后4组研究对象于2、5、和9 d各处死大鼠6只,应用ELISA法检测大鼠血清脂多糖结合蛋白(LBP)、肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)和Toll样受体4(TLR4)的表达;HE染色观察肝脏病理学改变并进行肝组织学活动指数(HAI)评分。结果:建模成功后2、5和9 d,OJ+NS组TNF-α、IL-6、TLR4、LBP水平均显著高于OJ+ω-3组,差异有统计学意义(P<0.05);OJ+NS组TLR4、TNF-α、IL-6水平显著高于SO+NS组,差异有统计学意义(P<0.05)。建模成功后2、5、9 d,根据HE染色及HAI评分,与OJ+NS组相比,OJ+ω-3组大鼠肝脏组织炎症状况改善,HAI评分明显降低(P<0.05)。结论:ω-3多不饱和脂肪酸可抑制梗阻性大鼠血清中LBP、TLR4、TNF-a、IL-6的表达,改善梗阻性黄疸大鼠炎症状况,从而达到调控炎症反应、保护肝脏的作用。 Objective: To investigate the effect of ω-3 polyunsaturated fatty acids on inflammatory response and liver function in obstructive jaundice rats. Methods: Seventy-two female Wisar rats were randomly divided into four groups(n=18): OJ+ω-3 group, OJ+NS group, SO+ω-3 group, SO+NS group. OJ+ω-3 group and OJ+NS group were used to construct the obstructive jaundice rat model by double ligation choledochotomy. SO+omega-3, SO+NS is only open for abdominal surgery. OJ+ω-3 group and SO+ω-3 group were given ω-3 fish oil fat emulsion by tail vein injection [5 mL/(kg·d)], OJ+NS group and SO+NS group were given the same volume physiological saline by tail vein injection. Six rats were executed in the 2 d, 5 d and 9 d, and ELISA was applied to detect the level of lipopolysaccharide binding protein(LBP), tumour necrosis factor-α(TNF-α), interleukin-6(IL-6) and toll-like receptors-4(TLR4); HE staining and histological activity index(HAI) were used to observe the histopathological changes. Results: In the 2 d,5 d and 9 d,compared with OJ+ω-3 group, TNF-α, IL-6, TLR4 and LBP levels of OJ+NS group were higher,the difference statistically significant(P0.05); In the 2 d, 5 d and 9 d, compared with SO+NS group,TLR, TNF-α and IL-6 levels of OJ+NS group were higher, difference was statistically significant(P0.05). According to HE staining and HAI scores, in the 5 d and 9 d, compared with OJ+NS group,the inflammation in the liver tissues of OJ+ω-3 group was improved, and the HAI scores was significantly reduced(P0.05). Conclusions: ω-3 polyunsaturated fatty acids can inhibit the expression of LBP, TLR4, TNF-α and IL-6, and improve the obstructive jaundice rat inflammatory conditions,thus can protect the liver.
