整合素αvβ6在甲状腺乳头状癌中的表达

Expression and roles of integrin αvβ6 in papillary thyroid carcinoma

目的:探究整合素αvβ6在甲状腺乳头状癌中的表达及其对肿瘤生物学行为的影响。方法:选取2014年11月—2015年9月60例PTC肿瘤及相应癌旁组织标本,免疫组化法检测整合素αvβ6的表达情况;在PTC细胞系TPC-1及B-CPAP中利用siRNA干扰整合素αvβ6表达,CCK8实验及Transwell侵袭实验分别检测PTC细胞增殖能力及侵袭能力的变化。结果:整合素αvβ6在癌旁组织中几乎不表达,在肿瘤组织中表达显著,且在伴有周围侵犯的肿瘤组织中表达高于不伴有周围侵犯的肿瘤组织。体外干扰αvβ6表达后,肿瘤细胞增殖能力及侵袭能力受到显著抑制。结论:在PTC组织中整合素αvβ6表达显著上调,上调的整合素αvβ6与肿瘤的侵袭浸润显著相关,干扰整合素αvβ6的表达可以显著抑制PTC细胞的增殖能力与侵袭能力。

Objective： To explore the expression and roles of integrin αvβ6 in papillary thyroid carcinoma（PTC）. Methods： Sixty PTC patients were selected from November 2014 to September 2015, the tumor tissues and the corresponding adjacent tissues were collected. Immunohistochemistry（IHC） was conducted to evaluate the expression of integrin αvβ6 in PTC tissues. Two PTC cell lines, TPC-1 and B-CPAP were selected and integrin αvβ6 was down-regulated by siRNA, followed by exploring the changes of proliferation and invasion using CCK8 and Transwell invasion assay,respectively. Results： IHC results showed that integrin αvβ6 negatively expressed in adjacent normal tissues, while it was significantly up-regulated in PTC tissues. Moreover, the integrinαvβ6 staining was much more stronger in PTC tissues with peripheral invasion compared with those without peripheral invasion. The proliferation and invasion capacities of PTC cells were markedly suppressed after integrin β6 silencing. Conclusion： The expression of integrin αvβ6 was significantly up-regulated in PTC tissues, and the increased αvβ6 expression was associated with tumor invasiveness. Silencing of integrin αvβ6 could markedly suppress the proliferation and invasion of PTC cells.
