加味七方胃痛颗粒对裸鼠原位移植胃癌中TFF3表达及ERK/NF-кB信号通路影响

Effect of Jiawei Qifang Weitong Granules on Orthotopic Transplantation of Gastric Cancer TFF3 and the Expression of ERK/NF-κB Signaling Pathway

目的：探讨加味七方胃痛颗粒对裸鼠原位移植胃癌三叶因子3（trefoil factor family3,TFF3）的表达及其细胞外信号调节激酶（extracellular signal-regulated kinase,ERK）/核因子кB（nuclear factor-кB,NF-кB）信号通路的调控机制。方法：复制裸鼠胃癌原位移植模型,随机分成正常组（即空白对照组）,氟尿嘧啶（5-fluorouracil,5-FU）组,5-FU＋加味七方胃痛颗粒组,加味七方胃痛颗粒组,每组15只。采用Western blot、RT-PCR检测加味七方胃痛颗粒对裸鼠原位移植胃癌中TFF3、NF-кB、ERK基因及蛋白表达的影响。结果：加味七方胃痛颗粒组TFF3、NF-кB、ERK m RAN及蛋白表达量较正常组降低（P〈0.05）,5-FU＋加味七方胃痛颗粒组TFF3、NF-кB、ERK m RAN及蛋白表达量较5-FU组降低（P〈0.05）。结论：加味七方胃痛颗粒可能通过抑制ERK/NF-кB信号通路参与下调裸鼠原位移植胃癌TFF3、NF-кB、ERK表达水平,从而达到抗癌作用。

Objective：To investigate the Jiawei Qifang Weitong（JWQFWT）Granules on orthotopic gastric cancer trefoil factor family3（trefoil factor family3,TFF3）and the expression of extracellular signal regulated kinase/nuclear factor-κB regulation signal pathway. Methods：The nude mice model of orthotopic transplantation of gastric cancer were randomly divided into blank control group,fluorouracil group,5-FU＋ modified JWQFWT Granules group,supplemented by the JWQFWT Granules group,each group of 15. To calculate the orthotopic transplantation of human gastric cancer in nude mice tumor inhibition rate by Western,blot,RT-PCR detection of modified JWQFWT Granules on TFF3 orthotopic gastric cancer,NF-κB,the expression of ERK gene and protein. Results：The modified JWQFWT Granules group TFF3,NF-κB,expression of ERK m RAN and protein were lower than those of blank control group（P〈0.05）,5-FU＋ with JWQFWT Granules group TFF3,NF-κB,expression of ERK protein and m RAN was lower than group 5-FU（P〈0.05）. Conclusion：The prescription of the JWQFWT Granules can effectively inhibit ERK/NF-κB signaling pathway involved in the down-regulation of orthotopic gastric cancer TFF3,NF-κB,ERK expression level,so as to achieve the anti-cancer effect.
