摘要
目的:研究痰浊阻窍AD大鼠在不同阶段的证候属性及证型演变。方法:采用"高糖高脂饮食+Aβ海马注射"方法制备痰浊阻窍证AD大鼠模型,将成模大鼠随机分为模型组、远志散组、远志散倍黄连组和远志散倍人参组,再根据干预时间不同分为Stage1、Stage2、Stage3 3个阶段。经相应药物干预4周后进行Morris水迷宫测试和western blot检测各组大鼠海马p T181蛋白的表达。结果:在Stage1远志散组效果最佳,其平均逃避潜伏期较模型组显著缩短(P〈0.01),穿越原平台次数增加(P〈0.01),有效区游泳路程延长(P〈0.05),海马中p T181蛋白表达降低(P〈0.05);在Stage2远志散倍黄连组效果最佳,其平均逃避潜伏期显著缩短(P〈0.01),穿越原平台次数增加(P〈0.01),有效区游泳路程延长(P〈0.05),海马中p T181蛋白表达显著降低(P〈0.01);在Stage3远志散倍人参组效果最佳,其平均逃避潜伏期缩短(P〈0.05),有效区游泳路程延长(P〈0.01),海马p T181蛋白的表达显著降低(P〈0.05)。结论:此AD模型大鼠在疾病发展过程中依次表现为痰浊阻窍、痰郁化火和痰盛气衰,早期偏于"痰",中期偏于"火",后期偏于"虚"。
Objective:To observe the nature and transformation on TCM syndromes in senile dementia rats(phlegm blockage syndrome)in different stages of disease. Methods:Senile dementia rats(phlegm blockage syndrome)were established with"high-fat and high-sucrose diet + injection of Aβ 25-35".Those with successful model establishment were randomized into control group,YuanzhiSan group,Yuanzhi San doubled Huanglian group and Yuanzhi San doubled Renshen group. Each of them was subdivided randomly into Stage1,Stage2 and Stage3 with corresponding herbal medicinal intervention. After the medicinal intervention for four weeks in each stage,changes of behavioristics in MWM test and p T181 in Western blot were observed. Results:The effect of of Yuanzhi San was better than that of other group in stage1,in which escape latency was decreased than control group(P〈0.01),the times across platform and swimming distance in the effective area were increased(P〈0.01,P〈0.05),the p T181 protein expression was reduced(P〈0.05).The effect of of Yuanzhi San doubled Huanglian was better in stage2,in which escape latency was decreased(P〈0.01),the times across platform and swimming distance in the effective area were increased(P〈0.01,P〈0.05),the p T181 protein expression was reduced(P〈0.01). The effect of of Yuanzhi San doubled Renshen was better in stage3,in which escape latency was decreased(P〈0.01),the swimming distance in the effective area was increased(P〈0.01),the p T181 protein expression was reduced(P〈0.05). Conclusion:In the course of disease development,the senile dementia rats appeared as phlegm blockage,stagnation of phlegm converting into fire,phlegmatic abundance and weakness of Qi. In the early stage,it is"phlegm",and it is"fire"in the middle stage,in the later period,it is partial to the"virtual".
