摘要
目的:探讨红芪总多糖(HPS)及红芪多糖HG-1(HHG-1)对大鼠骨关节炎(OA)的治疗作用及对局部炎症反应的影响。方法:将Wistar大鼠随机分为7组,采用木瓜蛋白酶复制OA动物模型,经HPS及HHG-1的干预,测定关节活动范围、关节滑膜病理变化及其中糖胺多糖、关节灌洗液中炎性细胞及炎症因子等指标,对HPS及HHG-1治疗OA的作用及机制进行研究。结果:HPS(关节给药1.05,0.35 mg)及HHG-1(关节给药0.45,0.15 mg)能明显扩大OA大鼠膝关节的伸展角度,改善关节病理变化,减小关节滑膜厚度,提高滑膜中糖胺多糖的含量;同时HPS及HHG-1可降低关节灌洗液中炎性细胞及IL-1β,IL-6,TNF-α水平(P<0.05)。结论:HPS及HHG-1对OA有一定的治疗作用,缓解局部炎症反应是其作用机制之一。
Objective: To investigate the therapeutic effect of Hedysari polysaccharides(HPS) and Hedysari polysaccharide HG-1(HHG-1) on osteoarthritis(OA) and the local inflammatory reaction in rats. Methods: The Wistar rats were randomly divided into seven groups,and the OA model was replicated with papain. After HPS and HHG-1 intervention,the range of joint activity,the pathological changes of synovial membrane and its content of glycosaminoglycan(GAG),inflammatory cells and inflammatory factors in synovial lavage fluid(SLF) were measured. Results: HPS(1. 05,0. 35 mg per joint) and HHG-1(0. 45,0. 15 mg per joint) can significantly expand the knee stretching angle,improve joint pathological changes,reduce synovial thickness,and increase the content of GAG in the synovial membrane of OA rats. At the same time,HPS and HHG-1 can decrease inflammatory cells and reduce the level of IL-1β,IL-6,TNF-α in SLF(P〈0. 05). Conclusion: HPS and HHG-1 had certain therapeutic effect on OA and one of the mechanisms is to alleviate the local inflammatory reaction.
