摘要
目的应用一测多评法同时测定蒲黄中4种成分的含量。方法采用UPLC法测定,以异鼠李素-3-O-新橙皮糖苷为内参物,计算各种成分的相对校正因子,并计算各成分的含量,实现一测多评。同时采用外标法测定4种成分的含量,与一测多评法测定结果进行比较,验证一测多评法的准确性。结果对羟基苯甲酸、香草酸和香蒲新苷的相对校正因子的平均值分别为4.043、2.167和0.851,采用校正因子计算的含量值与外标法实测值之间无显著差异,表明方法准确度高、重复性好。结论一测多评法可作为一个新的质量评价方法用于蒲黄中酚酸和黄酮类成分的含量测定。
Objective To assay the four components in Typhae Pollen by the method named quantitative analysis of multi-components by single marker( QAMS). Methods A UPLC method was developed to determine the contents of four components,and QAMS method was established when the isorhamnetin-3-O-neohesperidoside was used as internal referring substances. The relative correction factors( RCFs) between the three components and isorhamnetin-3-O-neohesperidoside were calculated and used to assay the contents of three components,which were compared with external standard method to verify the accuracy of the QAMS method.Results The mean values of the relative correction factors of p-hydroxybenzoic acid,vanillic acid and typhaneoside were 4. 043,2. 166 and 0. 851,respectively. There was no significant difference between the calculated values using the correction factors and the measured values of the external standard method,which indicated that the method was accurate and reproducible. Conclusion QAMS method can be used as a newquality evaluation method for the quality evaluation of phenolic acid and flavonoids in Typhae Pollen.
