黄芪总苷、赤芍总苷及丹参总酮对微波辐射后H9c2心肌细胞保护作用研究 被引量:6

摘要 目的探讨黄芪总苷、赤芍总苷及丹参总酮对微波辐射后H9c2心肌细胞活性氧及心肌酶谱改变的保护作用。方法 H9c2心肌细胞随机分为正常对照组(C)、辐射对照组(R)、1mg/l给药组(L)、3 mg/L给药组(M)和9 mg/L给药组(H)。黄芪总苷、赤芍总苷及丹参总酮采用1 mg/L,3 mg/L,9 mg/L 3个剂量预处理48 h后,采用平均功率密度为30 m W/cm^2的微波辐射H9c2心肌细胞,于辐射后30 min检测细胞中ROS含量、培养基中CK及LDH活性。结果辐射对照组心肌细胞中ROS含量和培养基中CK及LDH活性明显升高(P<0.05或P<0.01);黄芪总苷、赤芍总苷和丹参总酮组对上述指标改变呈现不同程度的改善作用;与辐射对照组比较,心肌细胞中ROS含量,黄芪总苷3个剂量给药组、赤芍总苷低剂量、丹参总酮低和高剂量给药组均明显降低(P<0.05或P<0.01);CK及LDH活性于黄芪总苷中、高剂量组及丹参总酮中剂量组明显降低(P<0.05)。结论 30 mW/cm^2微波辐射可引起H9c2心肌细胞CK、LDH活性及ROS含量升高;黄芪总苷、赤芍总苷及丹参总酮对上述指标改变的改善效果良好,其中黄芪总苷改善效果最佳,最佳有效剂量为3 mg/L。 Objective To investigate the protective effects of astragalosides,total paeony glycosides,and total tanshinones on improved abnormalities caused by microwave radiation of ROS content in myocardial cells and myocardial enzyme activity in the culture medium. Methods The H9 c2 myocardial cells were given microwave radiation with the average power density of 30 mW/cm^2,and were administered with1 mg/L,3 mg/L,9 mg/L Astragalosides,total paeony glycosides and total tanshinones 48 h before radiation. At 48 h before radiation and 30 min after radiation,drug cytotoxicity,the content of ROS in cells,activity of myocardial enzyme in culture medium were detected to determine the protective effects of Astragalosides,total paeony glycosides,and total tanshinones on microwave radiation-induced heart injury,to find the ingredients with best effect and the optimal dose for cellular level. Results 30 min after 30 mW/cm^2 microwave radiation,cardio myocytes ROS content and CK and LDH activities in culture medium were significantly increased( P 〈0. 05 or P 〈0. 01); Astragalosides,total paeony glycosides,and total tanshinones exhibit various degrees of improvement. Conclusions with the radiation control group,ROS content in myocardial cells,three Astragalosides groups with different dose,low dose total paeony glycosides group,total tanshinones groups of low and high dose were significantly lower( P〈0.05 or P〈0.01); myocardial enzyme activities was significantly lower in the Astragalosides groups of middle and high dose and in the middle dose total tanshinones group( P〈0.05). Conclusions 30 mW/cm^2 microwave radiation could cause increased activities of CK,LDH in H9 c2 cell culture medium and elevated levels of ROS in cells. The pretreatment using Astragalosides,total paeony glycosides and total tanshinones have a good effect on improving the above indexes. Among them,the Astragalosides was the best,and the optimal effective dose was 3 mg/L.
