人前列腺癌组织拓扑异构酶Ⅱα蛋白的表达水平及其对生化复发的预测意义 被引量:4

The expression of topoisomerase IIα in prostate cancer tissue and its significance in prediction of prostate cancer recurrence

摘要 目的研究拓扑异构酶Ⅱα在人前列腺癌组织中的表达水平,探讨其与生化复发的关系。方法回顾性分析我院30例人前列腺癌手术病理切片,其中16例Gleason评分≥7分,14例<7分。所有标本进行拓扑异构酶Ⅱa蛋白表达的免疫组织化学染色。随访患者,应用spss软件进行Kaplan-meirer生化复发的预后分析。结果拓扑异构酶Ⅱα蛋白在确诊为前列腺癌组织中26例呈阳性表达,阳性率为86.7%。拓扑异构酶Ⅱα的表达水平与Gleason评分存在一定相关性。当Gleason评分大于等于7分时,拓扑异构酶Ⅱα阳性表达水平较高;而Gleason评分小于7分时,拓扑异构酶Ⅱα阳性表达水平较低。两组间构成比存在显著性差异(P<0.05)。拓扑异构酶Ⅱα蛋白的表达水平与生化复发率存在一定相关性。拓扑异构酶Ⅱα在生化复发组的表达水平较高,其在未复发组的表达水平较低。结论人前列腺癌组织Gleason评分高,拓扑异构酶Ⅱα蛋白的表达水平高,生化复发率亦高,患者预后差。拓扑异构酶Ⅱα蛋白表达水平可以作为判断前列腺癌患者预后的生化指标。 Objective To investigate the expression of topoisomerase Ⅱα in prostate cancer tissue and explore its significance in prediction of prostate cancer recurrence. Methods Thirty prostate cancer specimens including 16 specimens whose Gleason score was more than 7 and 14 specimens whose Gleason score was less than 7, were retrospectively analyzed in our hospital. The expressions of topoisomerase Ⅱα in all the samples were detected by immunohistochemistry. All patients were followed up and the relationship between topoisomerase Ⅱα expression and recurrence free survival was analyzed by Kaplan-meirer. Results 86.7% of prostate cancer tissues had topoisomerase Ⅱα positive response. Gleason scores were correlated with topoisomerase Ⅱα level. When Gleason scores were over 7, the expression level of topoisomerase Ⅱα was high. When Gleason scores were below 7, the expression level was low.The expression level of topoisomerase Ⅱα in the biochemical recurrence free survival group are higher than that in the other group. Conclusion Topoisomerase Ⅱα can help us to confirm the diagnosis of prostate cancer. Topoisomerase Ⅱα expression was found to increase with the known prognostic marker Gleason score.
