基于核因子-κB通路研究复方柴芩退热颗粒对脂多糖介导细胞炎症模型的作用 被引量:1

Effect of compound Chaiqin Tuire granules on the cell model of lipopolysaccharide-mediated inflammation based on the nuclear factor-κB pathway

摘要 目的:研究复方柴芩退热颗粒对脂多糖介导炎症细胞模型的作用。方法:建立脂多糖介导RAW264.7细胞炎症模型,分为空白组,模型组,复方柴芩退热颗粒高、中、低浓度组,分别使用复方柴芩退热颗粒高、中、低剂量(21.12、10.56、5.28mg/m L)进行药物干预培养。酶联免疫法检测细胞培养上清液肿瘤坏死因子(TNF)-α、白介素(IL)-1β、IL-6及IL-10的含量。蛋白印迹法检测核转录因子(NF)-κB P-IκB表达水平。结果:复方柴芩退热颗粒高、中、低浓度组TNF-α、IL-1β及IL-6水平均显著低于模型组,差异有统计学意义(P<0.05),且呈浓度依赖性降低;IL-10水平均显著高于模型组,差异有统计学意义(P<0.05),但无浓度依赖性,其中复方柴芩退热颗粒高、中浓度组IL-10水平比较,差异无统计学意义(P>0.05)。复方柴芩退热颗粒高、中、低浓度组P-IκB表达水平均显著低于模型组,差异有统计学意义(P<0.05),且呈浓度依赖性降低。结论:复方柴芩退热颗粒具有显著抗炎作用,不仅能够显著降低LPS诱导细胞炎症模型P-IκB及TNF-α、IL-1β、IL-6的表达,同时还能升高IL-10的水平,其作用机制可能通过抑制IκB的磷酸化而阻断NF-κB信号通路有关。 Objective: To investigate the effect of compound Chaiqin Tuire granules on the cell model of lipopolysaccharide-mediated inflammation. Methods: RAW264. 7 cells were used to establish a model of lipopolysaccharide-mediated inflammation. The cells were divided into blank group,model group,and high-,medium-,and low-concentration compound Chaiqin Tuire granule groups,which were cultured with high-,medium-,and low-dose compound Chaiqin Tuire granules(21. 12,10. 56,and 5. 28 mg/ml,respectively). ELISA was used to measure the content of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),and inleukin-10(IL-10) in cell supernatant,and Western blot was used to measure the expression of nuclear factor-κB(NF-κB) P-IκB. Results: Compared with the model group,the high-,medium-,and low-concentration compound Chaiqin Tuire granule groups had significantly lower levels of TNF-α,IL-1β,and IL-6(P〈0.05)with reductions in a concentration-dependent manner and a significantly higher level of IL-10(P〈0.05) without concentration dependence,while there was no significant difference in the level of IL-10 between the high-and medium-concentration compound Chaiqin Tuire granule groups(P〈0.05). The high-,medium-,and low-concentration compound Chaiqin Tuire granule groups had significantly lower expression of P-IκB than the model group(P〈0.05) with reductions in a concentration-dependent manner. Conclusion: Compound Chaiqin Tuire granules have a marked anti-inflammatory effect and can significantly reduce the expression of P-IκB,TNF-α,IL-1β,and IL-6 and increase the level of IL-10 in a cell model of lipopolysaccharide-induced inflammation,possibly by inhibiting the phosphorylation of P-IκB and blocking the NF-κB signaling pathway.
