锶通过CaSR途径促进脂肪来源干细胞成骨分化的研究 被引量:1

摘要 目的从蛋白水平探讨锶(strontium,Sr)通过钙敏感受体(CaS R)诱导脂肪来源干细胞(ADSCs)成骨分化的机制。方法经体外分离、培养、鉴定ADSCs并进行成骨诱导,根据实验目的进行分组,诱导14 d后用Western blot方法检测细胞表面CaS R和Runt相关转录因子-2(Runx2)蛋白的表达。结果 Srcl2具有增强Runx2蛋白表达的作用,当加入CaS R特异性抑制剂NPS-2143时,可抑制细胞表面CaS R蛋白的表达,拮抗Sr^(2+)对Runx2蛋白的上调作用,削弱ADSCs成骨分化能力。结论 Sr2+可通过作用于CaS R发挥促进ADSCs成骨分化的效应。 Objective To explore the mechanism of strontium(Sr) in the induction of osteoblastic differentiation of adipose-derived stem cells(ADSCs) through the calcium-sensitive receptor(CaS R) gene at the protein level. Methods ADSCs were isolated, cultured, identified, placed under osteogenic induction, and grouped according to the purpose of the experiment. After 14 days of induction, the expression of CaS R and Runt-related transcription factor-2(Runx2) proteins were detected by Western blot. Results Srcl2 can enhance the expression of Runx2 protein. The addition of CaS R-specific inhibitor NPS2143 can inhibit the expression of CaS R protein on the cell surface, antagonize the up-regulation of Sr^2+ to Runx2 protein, and weaken the osteogenic differentiation ability of ADSCs. Conclusion Sr^2+ can promote the osteogenic differentiation of ADSCs by acting on the CaS R gene.
