幽门螺杆菌感染小鼠模型Th17细胞及IL-17表达变化的研究 被引量:3

摘要 目的:探讨Th17细胞和IL-17在幽门螺杆菌(Helicobacter pylori,H.pylori)感染小鼠模型脾脏和胃组织中的表达和意义。方法:35只H.pylori感染小鼠为实验组,35只正常小鼠为对照组,于感染后1、3、5、7、14、21、28 d分别处死5只实验组小鼠和5只对照组小鼠,流式细胞术检测H.pylori感染和正常对照小鼠脾脏Th17(CD4+IL-17+)细胞的百分率,ELISA方法检测H.pylori感染和正常对照小鼠胃组织中IL-17水平。结果:H.pylori感染14 d后,小鼠脾脏Th17细胞开始升高,21、28 d小鼠脾脏Th17细胞频数均显著高于对照组,且差异具有统计学意义(P<0.01);H.pylori感染7 d后,小鼠胃组织IL-17含量开始升高,21、28 d小鼠胃组织IL-17含量显著高于对照组(P<0.01)。结论:H.pylori感染小鼠脾脏Th17细胞数量增多,胃组织IL-17水平升高,Th17细胞和IL-17可能参与了H.pylori感染的免疫反应。 Objective: To investigate the percentage of Th17 cell in spleen and the level of IL-17 in gastric tissue in mouse models infected with Helicobacter Pylori(Hp). Methods: 35 mice(Hp+)were infected with Hp by intragastric administration of the bacterial suspension. Another 35 mice(Hp-)were chosen as the control group. Every 5 Hp+and 5 HP-mice were killed at day 1, 3, 5, 7, 14, 21,and 28, respectively. Th17 cell frequency in spleen of the infected mice and the control group was determined by the flow cytometry.The levels of IL-17 in the gastric tissues of the infected mice and the control group were determined by ELISA. Results: 14 days after HP infection, Th17 cells in the spleen of mice began to increase; and the Th17 cell frequency in the spleen of mice at 21 and 28 days were significantly higher than that of the control group, and the difference was statistically significant(P〈0.01). 7 days after HP infection, IL-17 content in gastric tissue of mice began to increase; both Th17 cell frequency in mice spleen and the level of IL-17 in mice gastric tissue of the Hp-infected mice at day 21 and 28 were significantly higher than those in the control group(P〈0.01).Conclusion: Hp infection can increase Th17 cell quantity in mice spleen and the IL-17 level in mice gastric tissue. Th17 cell and IL-17 may be involved in the Hp-induced immune responses.
