重庆地区羊传染性脓疱与病原菌混合感染的检测与分析 被引量:5

Detection and analysis of mixed infection of contagious ecthyma and pathogenic bacteria in goats of Chongqing

摘要 采集山羊口唇部痂皮,通过PCR检测、MDBK细胞培养、电镜观察、间接免疫荧光(IFA)检测对羊传染性脓疱病毒进行系统鉴定,同时,分离培养病灶部位主要病原菌,进行分子鉴定、致病性和药敏试验检测,初步分析病原特性。结果表明,共分离到1株羊传染性脓疱病毒,3种病原菌。通过对病原菌16SrDNA基因序列比对分析,发现其分别与多动物链球菌、金黄色葡萄球菌和溶血性曼氏杆菌同源性较高,在系统进化发育树中分别与其聚为一簇。致病性分析,发现多动物链球菌和溶血性曼氏杆菌对小鼠的致死率高达100%。3种病原菌主要侵染部位为小鼠心脏、肝脏和肺脏,能够引起肺脏和肝脏明显病变。药敏试验表明,3种菌均对舒巴坦、阿米卡星和培氟沙星等6种药物高度敏感。本研究首次从羊传染性脓疱病变部位分离到多动物链球菌、金黄色葡萄球菌和溶血性曼氏杆菌,为该病在重庆地区的流行特点和防治技术研究提供理论依据。 PCR detection, MDBK cell culture, electron microscopy and indirect immunofluorescence (IFA) were carried out for the systematic identification of contagious ecthyma in goats. The patho- gens were isolated and cultured, identified by molecular identification, pathogenic analysis and drug sensitivity test at the same time. The result showed that one virus strain and three kinds of patho- genic bacteria were successfully isolated. The pathogenic bacteria respectively were Streptococcus pluranimalium,Staphylococcus aureus ,Mannheimia haemolytica according to the 16S rDNA i- dentification. And each pathogenic bacteria formed a separate with the corresponding strains according to the system development of the evolutionary tree, respectively. The result of pathogenic analysis showed that the fatality rate in mice of Streptococcus pluranimalium and Mannheirnia haemolytica were as high as 100%. The main infection sites of the three pathogens were mouse heart,liver and lung, especially in the lungs and liver. The drug sensitivity test showed that all three strains were highly sensitive to six drugs,such as sulbactam,amikacin and pefloxacin,et al. Streptococcus pluranimalium , Staphylococcus aureus and Mannkeimia haernolytica were successfully isolated for the first time from the diseased region of contagious ecthyma in this study,it laid a foundation for the research of epidemic characteristics and control technique of contagious ecthyma in goat.
