藁苯内酯对自发性高血压大鼠AT1R基因表达的影响 被引量:5

摘要 目的:通过观察藁苯内酯对自发性高血压大鼠心肌组织AT1R基因表达的影响,探讨其影响高血压发生的作用机制。方法:将自发性高血压大鼠随机分为模型组、当归挥发油组(低、中、高)、藁苯内酯组、缬沙坦组,同周龄Wistar大鼠作为正常对照组,每组10只,模型组及正常对照组给予等量生理盐水灌胃。采用无创血压检测系统检测给药后1~4周各组大鼠血压;QT-PCR法检测AT1R m RNA表达量,Western Blot检测AT1R蛋白的表达水平。结果:与模型组比较,各给药组血压下降差异显著(P〈0.05),藁本内酯组AT1R的m RNA及蛋白表达水平均下调,差异显著或极显著(P〈0.05或P〈0.01)。结论:当归可能通过其有效成分藁苯内酯对血压有干预作用,可能通过AT1R靶点发挥作用。 Objective: To study the influence of ligustilide on AT1 R gene expression in spontaneously hypertensive rats(SHR).Methods: All the rats were divided into Radix Angelicae Sinensis group,model control group,ligustilide group and valsartan group with weekly-old Wistar rats as normal control group. The systolic blood pressure of the tail in different groups before and after treatment was measured. After 4 weeks,the m RNA level of AT1 R was detected by QT-PCR,and the protein expression of AT1 R was tested by Western Blot. Results: Ligustilide could reduce blood pressure level in SHR. The m RNA and protein expression of AT1 R was significant increased in Radix Angelicae Sinensis group and ligustilide group(P 0.01 or P 0.01). Conclusion:Ligustilide can reduce the blood pressure through down regulation the expression of AT1 R gene.
