摘要 目的:观察电针预处理对换笼失眠大鼠睡眠觉醒的影响,探讨电针预处理干预换笼失眠大鼠睡眠觉醒的机制。方法:将160只大鼠随机分为对照组、模型组、电针组和假电针组,每组分为5个亚组,分别为换笼后0 min、30 min、2 h、6 h 4个亚组和睡眠监测亚组,每个亚组各8只。观察4组大鼠电针干预前后睡眠觉醒的变化、换笼后各时点血清激素变化及换笼后24 h HPA轴多巴胺受体变化情况。结果:同对照组相比,换笼后模型组和假电针组表现为日间wake时间延长(P<0.05,P<0.01)、NREM睡眠减少(P<0.05);电针组睡眠觉醒量与对照组比较,差异无统计学意义(P>0.05)。对照组与模型组大鼠外周血激素水平比较,模型组、假电针组与电针组比较,差异均无统计学意义(P>0.05)。换笼后24 h模型组大鼠HPA轴的多巴胺D1受体和多巴胶D2受体表达水平显著降低,电针组HPA轴的多巴胺D1受体和多巴胺D2受体表达水平升高。结论:电针预处理能够干预换笼失眠大鼠睡眠觉醒,可能通过干预HPA轴多巴胺受体发挥作用。 Objective: To observe the preconditioning effect of electro-acupuncture( EA) on sleep-wake in rats with insomnia due to cage change,and to explore the mechanism of EA preconditioning. Methods: A total of 160 SD rats were randomized into the control group,the model group,the EA group and the sham-EA group,each group were sub-divided into five sub-groups including 0 minute after cage change,30 minutes after cage change,2 hours after cage change,and 6 hours after cage change,as well as the sub-group for monitoring sleep,with 8 rats in each sub-group. The changes of sleep-wake in the four groups were observed before and after the EA intervention. The changes of serum hormone after the cage change and the expression of dopamine receptors in the HPA axis 24 hours after the cage change were also observed. Results: In the model group and the sham-EA group,the wake duration were significantly prolonged( P〈0. 05,P〈0. 01),and NREM sleep was significantly decreased( P〈0. 05) during the daytime compared to those of the control group after the cage change. There was no statistical difference between the EA group and the control group in terms of the sleep-wake amounts( P〉0. 05). In terms of serum hormone,there was no significant difference between the control group and the model group( P〉0. 05); also there were no significant differences among the model group,the EA group and the sham-EA group( P〉0. 05). After the cage change,the expressions of D1 and D2 in the HPA axis were significantly decreased in the model group,whereas they were significantly increased in the EA group. Conclusion: EA preconditioning can intervene the sleep-wake in rats with insomnia due to cage change,and its function may be related to intervening the expressions of dopamine receptors in the HPA axis.
