摘要
目的探讨NOD样受体家族蛋白3(NOD-like receptor protein 3,NLRP3)与牙髓炎的相关性。方法选取2016年3月至2017年4月于南京市口腔医院治疗患者拔除的55颗第三磨牙为研究对象,其中正常牙齿15颗(正常组),龋坏牙齿20颗(龋坏组),牙髓炎牙齿20颗(牙髓炎组)。采用免疫组化染色检测NLRP3的表达,采用蛋白质印迹法检测NLRP3和白介素-1β(interleukin-1β,IL-1β)蛋白相对表达量,采用逆转录聚合酶链反应检测NLRP3和IL-1βmRNA的表达。结果免疫组化染色显示:牙髓炎组NLRP3表达明显强于龋坏组和正常组,龋坏组NLRP3表达明显强于正常组。牙髓炎组NLRP3 mRNA和蛋白相对表达量均明显高于龋坏组和正常组(P_均<0.05),龋坏组NLRP3 mRNA和蛋白相对表达量均明显高于正常组(P_均<0.05);牙髓炎组IL-1βmRNA和蛋白相对表达量均明显高于正常组(P_均<0.05),但与龋坏组比较差异均无显著性(P_均>0.05);龋坏组IL-1βmRNA和蛋白相对表达量均明显高于正常组(P_均<0.05)。NLRP3 mRNA与IL-1βmRNA的表达呈正相关(r_(正常组)=0.312,r_(龋坏组)=0.301,r_(牙髓炎组)=0.287;P_均<0.05),NLRP3与IL-1β的蛋白表达呈正相关(r_(正常组)=0.331,r_(龋坏组)=0.280,r_(牙髓炎组)=0.282;P_均<0.05)。结论 NLRP3在牙髓炎组织中高表达,其可能参与了牙髓的炎性反应。
【Abstract】 ObjectiveTo investigate the correlation of the pyrin domain containing NOD like receptor family protein 3 (NLRP3) and pulpitis. MethodFrom March 2016 to April 2017, 55 teeth of third molars which needs extractions for treatment were selected in Nanjing Stomatological Hospital, normal of 15 teeth (normal group), caries of 20 teeth (caries group,) pulpitis of 20 teeth (pulpitis group), immunohistochemistry was performed to detect the expression of NLRP3 protein, NLRP3 and interleukin-1β (IL-1β) protein expression was detected by Western blotting assay, NLRP3 and IL-1β mRNA expression was detected by reverse transcriptase polymerase chain reaction (RT-PCR). ResultImmunohistochemical staining showed that NLRP3 expression in pulpitis group was signi?cantly stronger than that in caries group and normal group, and NLRP3 expression in caries group was signi?antly stronger than that in normal group. The expression levels of NLRP3 mRNA and protein in pulpitis group were sign?cantly higher than those in caries group and normal group (Pall〈 0.05), the expression levels of NLRP3 mRNA and protein in caries group were signicantly higher than those in normal group (Pall〈0.05). The expression levels of IL-1β mRNA and protein in pulpitis group were signi?cantly higher than those in normal group (Pall〈 0.05), but compared with caries group had no signi?cant di?erence (Pall〉 0.05). The expression levels of IL-1β mRNA and protein in caries group were signi?cantly higher than those in normal group (Pall〈0.05). The expression of NLRP3 mRNA was positively correlated with the expression of IL-1β mRNA (rnormal group = 0.312, rcaries group = 0.301, rpulpitis group =0.287, Pall〈0.05), the protein expression of NLRP3 was positively correlated with the protein expression of IL-1β (rnormal group =0.331, rcaries group = 0.280, rpulpitis group = 0.282, Pall〈 0.05). ConclusionThe expression of NLRP3 in pulpitis tissue are increased, may participate in the inflammatory reaction of pulp.
