金钗石斛芽组织培养基质的筛选 被引量:3

摘要 探明金钗石斛芽组织培养的最佳诱导培养基、生根培养基、移栽基质等,建立金钗石斛芽组织培养技术体系,为金钗石斛资源保护及可持续利用提供有效途径,以金钗石斛芽的诱导率、芽的增殖倍数、芽长势、芽生根率、根条数、组织苗的成活率和长势为指标,进行了金钗石斛芽组织培养最佳诱导培养基、生根培养基、移栽基质的筛选试验。结果表明:金钗石斛芽的最佳诱导培养基为MS+6-BA 0.5mg/L+NAA0.1mg/L+3%蔗糖+0.3%琼脂,增殖倍数达3.37倍,且芽多、长势壮;最佳生根培养基为1/2MS+IBA0.8mg/L+NAA 0.5mg/L+水蛋白400mg/L+活性炭8g/L+3%蔗糖+0.3%琼脂,生根率达33.3%;最适移栽基质为锯木屑,移栽150d时成活率、生长量分别为46%和4.4cm。 In order to explore the optimal inducing medium, rooting medlum and transplanting medium for Dendrobium nobile buds tissue culture, establish the tissue culture technology system, and provide an effective path for D. nobile protection and sustainable utilization,screening experiments of optimal inducing medium, rooting medium and transplanting medium for D. nobiZe buds tissue culture were conducted to explore the Bud performances of inducing rate, proliferation rater growth vigor, growth rooting rate, number of roots, seedling survival rate and growth vigor. Results: The optimal inducing medium for D. nobile buds was MS+6-BA 0.5 mg/L+NAA 0.1 mg/L+3% sugar+0.3% agar, with the proliferation rate of 3.37, multiple buds and strong growth vigor; The optimal rooting medium was 1/2MS+IBA 0.8 mg/L4-NAA 0. ,5 mg/L+400 mg/L aquaporin+8 g/L active carbon+3% sugar+0.3% agar,with the rooting rate of 33. 3%; The optimal transplanting medium was sawdust, the survival rate and growth amount 150 d after transplanted were respectively 46G and 4.4 cm.
