摘要
目的探讨瑞舒伐他汀对脑出血大鼠血清白介素-6(IL-6)、脑组织水通道蛋白-4(AQP-4)与金属蛋白酶-9(MMP-9)表达水平和神经功能的影响。方法选取90只雄性大鼠作为研究对象,按随机数字表法将其分为A组,B组和C组,每组30只,A组大鼠未做任何处理,B组和C组大鼠均建立脑出血模型,B组大鼠予以0.9%氯化钠溶液治疗,C组大鼠予以瑞舒伐他汀治疗,分别记录比较3组大鼠不同时间段其血清IL-6、AQP-4和MMP-9表达水平以及神经功能情况。结果 A组大鼠在治疗后6 h、12 h、1 d、3 d和5 d时其IL-6、AQP-4与MMP-9表达水平与神经功能评分均无明显变化(P>0.05),而B、C组大鼠各时间点其IL-6、AQP-4和MMP-9表达水平均明显高于A组,除去治疗后6 h外,其余各时间点B组IL-6、AQP-4与MMP-9表达水平均显著高于C组(P<0.05);除去治疗后6 h和12 h外,B、C组大鼠其神经功能评分均较A组升高,且B组升高程度尤为明显(P<0.05)。结论 L-6、AQP-4与MMP-9均与大鼠脑出血后疾病的进展均有着密切关系,而瑞舒伐他汀可能通过降低IL-6、AQP-4与MMP-9的表达水平从而改善其神经功能。
Objective To investigate the effects of rosuvastatin on the serum levels of interleukin-6(IL-6),the expression of aquaporin-4(AQP-4) and metalloproteinase-9(MMP-9) in brain tissue as well as neurological function. of rats with intracerebral hemorrhage(ICH). Methods Ninety male rats were randomly divided into group A(n = 30),group B(n = 30) and group C(n = 30). The rats in group A were not treated. Cerebral hemorrhage models were established in both group B and group C. And the rats in group B and C were treated by 0. 9% Na Cl solution and rosuvastati,respectively. The serum levels of IL-6,expression of brain tissue AQP4 and MMP-9 and neurological function of rats at different time points in the three groups were recorded and compared. Results There were no significant changes in levels of IL-6,expression of brain tissue AQP4 and MMP-9 in brain tissue as well as neurological function of rats at 6 h,12 h,1 d,3 d and 5 d after treatment in group A. However the levels of IL-6,expression of brain tissue AQP4 and MMP-9 and neurological function of rats at different time points in group B and group C were significantly higher than those in group A(P〈0. 05). Moreover,the levels of IL-6,expression of AQP4 and MMP-9 in brain tissue as well as neurological function of rats at 12 h,1 d,3 d and 5 d after treatment in group B were significantly higher than those in group C(P〈0. 05). In addition,the scores of neurological function of rats at 1 d,3 d and 5 d after treatment in group B and group C were higher than those in group A,in which those in group B were more significant(P〈0. 05). Conclusion IL-6,AQP4 and MMP-9 are closely related to the progression of the disease after intracerebral hemorrhage in rats. Rosuvastatin may improve the neurological function by reducing the levels of IL-6,the expression of AQP4 and MMP-9 in brain tissue of rats with intracerebral hemorrhage.
