摘要
目的:采用紫外吸收光谱法研究奥扎格雷与牛血清白蛋白(BSA)之间的相互作用。方法:将奥扎格雷与BSA溶液按一定比例混合,考察不同pH、温度、反应时间、药物用量对其相互作用的影响,得出最佳实验条件。在此条件下,利用奥扎格雷和BSA的紫外吸收图谱和Lineweaver-Burk双倒数曲线模型得出两者的结合常数。结果:奥扎格雷与BSA的结合常数K=(6.6±0.6)×10~4L·mol^(-1),体系的吸光度与浓度成正比,回归方程为A=0.301+3.72×10~4c(r=0.999 5),线性范围为1.98×10^(-6)~3.05×10^(-5)mol·L^(-1)。使用本方法测得BSA制剂中BSA的平均百分质量分数为98%,平均回收率为(99.0±0.8)%,相对标准偏差为0.16%。结论:紫外吸收光谱法操作简便、灵敏度高、选择性好,可用于BSA制剂中BSA含量的测定。
Objective: To study the interaction between ozagrel and bovine serum albumin by ultraviolet absorption spectroscopy. Methods: Combining different proportions of ozagrel and bovine serum albumin,effects of different conditions such as pH,reaction temperature,reaction time and the concentration of the ozagrel on their interaction was investigated. The optimal experimental condiiton was obtained. Under the optimal experimental condition,according to the UV spectra of ozagrel and bovine serum albumin and Lineweaver-Burk double reciprocal curve,the binding constant was calculated. Results: The binding constant K was( 6. 6 ± 0. 6) × 10~4 L·mol^(-1). Under the optimal conditions,the absorbance of system was linear with the concentration of BSA,a linear regression equation was obtained as A = 0. 301 + 3. 72 × 10~4 c( r = 0. 999 5) in the range of 1. 98 ×10^(-6)~ 3. 05 × 10^(-5) mol·L^(-1). The average percentage of mass fraction of BSA was 98%. The average recovery rate was( 99. 0± 0. 8) %,and the RSD was 0. 16% respectively. Conclusion: The method was simple for determining the content of BSA with high sensitivity and selectivity.
