流感病毒H5N1的小RNA表达谱研究

Study on the Small RNA Expression Profile of Influenza Virus H5N1

H5N1病毒是一种对人高致死性的流感病毒亚型。为探索H5N1病毒高毒性原因,对H5N1病毒感染细胞开展了病毒源小RNA研究。结果显示,H5N1感染细胞中病毒源小RNA比例高;H5N1病毒小RNA的分布没有末端偏好;病毒源小RNA中存在1条定位于颈环结构的sRNA;H5N1病毒NS1蛋白5个关键氨基酸缺失可能与H5N1、H1N1小RNA谱的差异有关。它们为揭示H5N1病毒的高致死性机制提供了重要遗传基础,为高致死性流感的预防提供了参考靶点。

H5 N1 virus is a highly lethal subtype of human influenza virus.In order to explore the causes of H5 N1＇s high virulence,an analysis of small RNA profile from virus was carried out against cells infected by H5 N1.The results showed high proportion of small RNA in H5 N1 infected cells and the sRNAs derived from H5 N1 do not have the preference to locate on the terminal of the RNA segment of the virus.There was 1 sRNA located in a stem-loop structure.The difference of small RNA frofiles between H5 N1 and H1 N1 may relate to a deficiency of 5 amino acids of H5 N1.These results provide an important genetic basis for revealing the highly lethal mechanism of H5 N1 virus,and provide a potential target for the prevention of highly lethal influenza.
