摘要
目的:建立冬虫夏草中尿苷、鸟苷、腺苷3种核苷类物质的HPLC含量测定方法,并对市场上现有冬虫夏草正品、伪品和代用品中的3种核苷类物质含量进行分析。方法:色谱柱采用Zorbax Eclipse Plus C18柱(250mm×4.6mm,5μm),以甲醇-水为流动相,梯度洗脱(0~10min 5%甲醇,11~15min 7%甲醇,16~20min 15%甲醇,21~25min 30%甲醇),检测波长260nm,流速1.0ml/min,柱温30℃,进样量20μl。结果:3种核苷类物质含量在1~25μg/ml浓度范围内与峰面积线性关系良好(r=0.999 9),精密度、稳定性和加样回收率指标均符合要求。27个不同品种冬虫夏草中的尿苷和鸟苷平均含量正品大于伪品和代用品。结论:正品冬虫夏草中的尿苷和鸟苷含量明显高于伪品和代用品,可以为其正品鉴定提供参考依据。
Objective:To establish a HPLC method for the determination of three kinds of nucleosides(uridine,guanosine and adenosine),so as to analyze the contents of the three nucleosides in authentic Cordyceps sinensis,its counterfeit products and subtitutes on the market.Methods:Chromatography separation was performed on the Zorbax Eclipse Plus C18 column(250 mm×4.6 mm,5μm)with methanol-water as mobile phase in a gradient elution(0-10 min 5% methanol,11-15 min 7%methanol,16-20 min 15% methanol,21-25 min 30% methanol).Detection wavelength was 260 nm,flow rate was 1.0 ml/min,column temperature was 30℃,and injection volume was 20μl.Results:The concentrations and the peak areas of the three kinds of nucleosides within the range of 1-25μg/ml was in good linearity(r=0.999 9).The methodological indicators of presicion,stability and recovery rate all satisty the required demands.The average contents of uridine and guanosine in authentic C.sinensis were higher than those in the counterfeit produces and substitutes.Conclusion:The contents of uridine and guanosine in authentic C.sinensis are obviously higher than those in the counterfeit products and substitutes.It may provide reference for the identification of authentic C.sinensis.
