基于晋汾52和晋谷21解析谷子NBS类型抗病基因变异 被引量:2

摘要 [目的]为明确抗病品种晋汾52与其辐射诱变的感病品种晋谷21的NBS抗病基因之间存在的变异差异及引起抗性差异的原因。[方法]基于前期全基因组测序筛选出的6个抗病基因为基础,克隆了晋汾52和晋谷21相应的抗病基因,并利用生物信息学软件比对抗病基因序列,预测其蛋白结构及系统进化分析。[结果]筛选出2个变异位点差异较大的基因,Si028867和Si033294,经分析推测两个品种之间抗性差异主要由非同义突变引起。[结论]NBS类抗病基因之间的非同义突变可能是导致两个品种之间抗病性存在差异的原因。该研究结果对谷子抗病分子辅助育种具有一定理论指导意义。 [Objective]Present study was aimed to clarify the genetic variation and the potential causes of NBS genes in disease resistance of both resistant variety,Jinfen 52,and susceptible mutant strain,Jingu21 of foxtail millet.[Methods]According to previous genome sequencing outcomes,six resistant genes were cloned,and the disease resistance gene sequences was compared by bioinformatic software.Protein structure of cloned genes was predicted and phylogenetic analysis was performed.[Results]Two genes with large variations in mutation site,Si028867 and Si033294,were detected.It was speculated that the resistance difference between the two cultivars were mainly induced by nonsynonymous mutations.[Conclusion]The non-synonymous mutation may lead to resistance differences in Jinfen52 and Jingu21.The results will provide theoretical guiding for molecular assisted breeding of millet varieties with disease resistance.
