摘要
羟氯喹作为传统抗疟疾药物,目前主要用于风湿免疫性疾病的治疗,并且还具有抗肿瘤活性,其作用机制包括:影响溶酶体功能,抑制细胞自噬,调节免疫,影响相关蛋白及通路,抑制炎症反应,影响内涵体功能,通过线粒体途径诱导细胞凋亡以及化疗增敏等。本文拟对羟氯喹的抗肿瘤机制及其抗肿瘤临床试验进行综述,以期为进一步的研发和应用奠定基础。
As a traditional anti-malaria drug,hydroxychloroquine is currently used for the treatment of rheumatic autoimmune diseases and it also has antitumor activity and the mechanisms include affecting lysosomal function,inhibiting autophagy,regulating immune,affecting related proteins and pathways,inhibiting inflammatory,influencing the function of endosome,inducing apoptosis through mitochondria pathway and sensitizing chemotherapy. In this paper, the anti-tumor mechanisms of hydroxychloroquine and its clinical trials are reviewed in order to lay a foundation for further research and application of hydroxychloroquine.
