摘要
目的:观察百合地黄汤对慢性不可预见性应激抑郁大鼠模型血清炎症因子及其海马5-HT表达水平的影响,探究其抗抑郁的作用机理。方法:将60只SD大鼠随机分为正常组、模型组、百合地黄汤低剂量组、百合地黄汤高剂量组、湘百合地黄汤组、氟西汀组,每组10只。采用孤养加慢性温和不可预知性应激的方法建立慢性应激抑郁大鼠模型,造模时间为28 d,造模结束后,通过旷场实验,强迫游泳实验评价大鼠的抑郁样行为,ELISA法检测各组大鼠血清抗炎因子IL-1β的水平变化,免疫组化法检测大鼠海马神经递质5-HT的表达。结果:模型组大鼠的自主活动次数明显少于正常组,强迫游泳不动时间明显长于正常组,血清IL-1β含量明显高于正常组,海马神经递质5-HT表达明显低于正常组(P<0.01);百合地黄汤高剂量组及湘百合地黄汤组大鼠的自主活动次数明显多于模型组、强迫游泳不动时间明显短于模型组,血清IL-1β含量明显低于模型组,海马神经递质5-HT表达明显高于模型组(P<0.01);百合地黄汤高剂量组大鼠的行为学、血清IL-1β含量及海马5-HT表达情况与湘百合地黄汤组比较,差异无统计学意义(P>0.05)。结论:百合地黄汤对抑郁模型大鼠抗抑郁疗效显著,抑制慢性应激抑郁模型大鼠血清IL-1β含量升高,提高其海马5-HT表达水平,可能是其抗抑郁作用机理之一;百合地黄汤方中选用湖南龙山百合与选用江西百合比较,对其抑郁作用影响无明显差异。
Objective: To observe the effeet of Baihe Dihuang Decoction on the changes of serum anti-in- flamlnatory cytokines and neurotransmitter in rats with chronic unpredictable stress (CUMS) depression and to ex- plore its antidepressant mechanism. Methods: The 60 SD rats were randomly divided into blank control group, depression model group, low dose Baihe Dihuang Decoction group, high dose Baihe Dihuang Decoction, Xiang- Baihe Dihuang Deeoction group and fluoxetine group. The rats model of chronic stress depression was estab- lished by solitary and chronic mild unpredietable stress. The levels of serum anti-inflammatory cytokines IL- l lSwere detected by ELISA. The levels of hippocampal neurotransmitters 5-HT expression were detected by Im- munohistochemistry. Results: The in the model group showed lower number of grid crossing, longer immobility time inforeed swimming test, higher levels of IL-1β and lower expression of neurotransmitter 5-HT in hippocam- pus than the normal group (P〈0.01). High dose Baihe Dihuang Decoction and Xiang-Baihe Dihuang Decoction group showed higher number of grid crossing, less immobility time inforced swimming test, lower levels of IL-Iβ and higherer expression of neurotransmitter 5-HT in hippocampus than the model group (P〈0.01). Ther was no significant difference on between the high dose Baihe Dihuang Decoction and Xiang-Baihe Dihuang Decoction group on rat behavior, levels of IL-1β and expression of neurotransmitter 5-HT in hippocampus (P〉0.05). Con- clusion: Baihe Dihuang Decoc, tion has significant antidepressant effect on depression model rats. It may be one of the mechanisms of its antidepressant effect by effeetively inhibiting the increase of serum IL-1β level and in- creasing the expression of 5-HT in hippocampus of chronic stress depression model rats; The selection f oHu- nan Longshan lily compared with the selection of Jiangxi lily in the Baihe Dihuang Decoction shows no signifi- cant difference on its antidepressant effect.
