摘要
目的:研究万寿菊叶黄素对乳腺癌MCF-7细胞株增殖和凋亡的影响。方法:选用对数生长期的乳腺癌MCF-7细胞为研究对象。按随机分配原则,将其分为六组:叶黄素(lutein)处理组浓度分别为5、10、20、40、50μg/m L,未处理组作为对照组。用不同浓度的叶黄素分别处理MCF-7细胞,并培养12、24、48h,噻唑蓝(MTT)法计算乳腺癌MCF-7细胞的增殖抑制率;流式细胞法检测MCF-7乳腺癌细胞凋亡率。结果:乳腺癌MCF-7细胞在相同的培养条件下,随着叶黄素浓度升高,叶黄素对人乳腺癌MCF-7细胞的抑制作用及凋亡率显著增加。差异有统计学意义(P<0.05)。即相同的作用时间,用药浓度越大时,对MCF-7细胞的抑制作用和凋亡作用越强。用相同浓度的叶黄素处理MCF-7细胞,随着细胞的培养时间逐渐增加,细胞的增值抑制率及凋亡率呈明显上升趋势。且差异有统计学意义(P<0.05)。结论:万寿菊叶黄素对MCF-7乳腺癌细胞的增殖有抑制作用,并能诱导乳腺癌MCF-7细胞凋亡。
Objective To study the effect of marigold lutein on proliferation and apoptosis of human breast cancer McF-7 cell lines. Methods: The McF-7 cells of breast cancer in logarithmic growth period were studied. According to the principle of random distribution, it was divided into six groups: lutein concentration was 5, 10, 20, 40, and 50 respectively. The untreated group was taken as the control group. After treating McF-7 cells for 12, 24 and 48 hours with lutein of different concentrations, the inhibition rate of McF-7 cell prolif- eration was calculated by thiazolan (MTT) method. The apoptosis rate of MCF-7 cells was detected by flow cytometry. Statistical methods were used to analyze whether there were differences in the proliferation inhibition rate and apoptosis rate of McF-7 cells in the treatment group with different action times and different concentration of lutein at the same concentration. Results: Breast cancer MCF - 7 cell in the same incubation time conditions, with rising lutein concentration , lutein in human breast cancer MCF - 7 cell proliferation inhibition and apoptosis rate increased significantly. The difference was statistically significant (P〈0.05). That is, the same action time, the higher the drug concentration, the stronger the proliferation inhibition and apoptosis of McF-7 ceils. MCF - 7 cell in the same concentration of lutein processing conditions, gradually increased along with the cell incubation time , the inhibition rate of cell's appreciation and apopto- sis rate showed a trend of obvious rise. The difference was statistically significant (P〈0.05). Conclusion: Marigold lutein can inhibit the proliferation of McF-7 breast cancer cells and induce the apoptosis of McF-7 cells.
