探讨透骨草总生物碱对人乳腺癌MDA-MB-231细胞生长、迁移侵袭的作用及其机制研究

To investigate the effect and mechanism of total alkaloids of Tougucao on human breast cancer MDA-MB-231 cell growth,migration and invasion

目的探讨透骨草总生物碱(TTAT)对MDA-MB-231细胞生长、迁移和侵袭的作用,并对其机制进行研究。方法实验分为正常组、实验组和阳性对照组,体外培养MDA-MB-231细胞,MTT法检测TTAT对细胞生长、增殖的影响,Transwell实验和细胞划痕实验检测TTAT对人乳腺癌MDA-MB-231细胞侵袭转移能力的作用,Western Blot法测定不同浓度TTAT处理人乳腺癌后,转移相关蛋白金属基质蛋白-2(MMP2)、MMP9、血管内皮生长因子(VEGF)的表达。结果TTAT对人乳腺癌MDA-MB-231细胞的生长、迁移和侵袭有明显的抑制作用,并具有一定的量效关系,且转移相关蛋白MMP2、MMP9、VEGF的表达均下调。结论透骨草总生物碱能够抑制人乳腺癌MDA-MB-231细胞的增殖、迁移和侵袭能力,其机制可能与抑制MMP2/9、VEGF的表达以及二者之间的协同作用有关。

Objective To investigate the effect of the total alkaloids of Tougucao（ TTAT） on the migration and invasion of MDA-MB-231 cells,and to study its mechanism. Methods The experiment was divided into normal group,experimental group and positive control group. Transwell assay and cell scratch test were used to detect the effect of TTAT on invasion and metastasis of human breast cancer MDA-MB-231 cells. The expressions of metastasis related proteins,metal matrix proteins-2（ MMP2）,MMP9 and vascular endothelial growth factor（ VEGF） were measured by Western blot method after different concentrations of TTAT were used to treat human breast cancer. Results TTAT can inhibit the migration and invasion of human breast cancer MDA-MB-231 cells,and has a dose effect relationship. The expression of related proteins MMP2,MMP9 and VEGF were down regulated. Conclusion The total alkaloids of tougucao（ TTAT） can inhibit the proliferation,migration,invasion of human breast cancer cell line MDA-MB-231,the mechanism may be related to the inhibition of the expression of MMP2/9 and VEGF,and the synergistic effect between the two.
