摘要
目的:使用生物信息学的方法,分析肺癌细胞(A549)经慢病毒感染后,过表达超保守区基因与正常肺癌细胞之间的差异基因,并进行基因功能和信号通路分析,为肺癌研究提供差异基因。方法:在公共数据库(GEO)中下载6个肺癌细胞差异基因表达阵列数据进行生物信息学分析,筛选出相关的差异基因。使用DAVID和STRING数据库对差异基因表达进行基因功能、信号通路和蛋白互作用分析。结果:共得到230个差异基因,其中217个表达上调,13个表达下调;GO富集分析显示差异基因主要定位于细胞膜外,参与血管内皮生长因子的调节;KEGG信号通路分析显示差异基因主要表现在化学致癌上;蛋白互作网络显示主要的相关蛋白是CFH、MUC5B、PTGS2、LRRK2。结论:本研究从生物信息学方法对数据进行处理,在基因水平上表明肺癌细胞过表达超保守区后存在差异基因,为基础研究筛选出差异,进而服务于临床。
Objective: Using bioinformatics method analysis of lung cancer cells( A549) lentivirus infection ultraconservative district between normal lung cancer cell different genes and analysis gene function、signal pathway for genetic variations. Methods: The dates of whole genomic expression profiles got from the lentivirus infection ultraconservation district lung cancer cell and normal lung cancer cell were obtained from GEO database and identified differential expressed genes of lung cancer cell. The differential expressed genes were characterized by gene ontology,KEGG pathway and protein-protein interaction network analysis. Results: A Total of 230 different genes( 217 upregulated and 13 downregulated) were found in GEO series. GO analysis showed that differences of enrichment of gene mainly located in cell membrane,participate in the regulation of vascular endothelial growth factor; KEGG pathway analysis showed that genetic variations in chemical carcinogenesis; Protein interaction network shows the main related proteins is CFH,MUC5B,PTGS2 et. al. Conclusion: This study using bioinformatics method for data processing,which shows that after lung cancer cells express ultraconservative district difference gene in the genetic level. Study screened the differences on the basis of the method and serviced clinical.
