大口黑鲈抗菌肽提取及抑菌活性初步研究 被引量:2

摘要 用5%乙酸提取大口黑鲈(Micropterus salmoniodes)肝脏、脾脏、皮肤、粘液和卵组织中的抗菌肽,采用考马斯亮蓝G-250法测定大口黑鲈各组织的抗菌肽蛋白质含量,采用纸片琼脂扩散法检测其对大肠杆菌(Escherichia coli)、嗜水气单胞菌(Aeromona hydrophila)、金黄色葡萄球菌(Staphylococcus aureus)的抑菌效果。结果表明:大口黑鲈各组织提取物的蛋白质含量依次为卵﹥皮肤﹥肝脏﹥脾脏﹥粘液,卵2.515 mg·L^(-1),皮肤0.643 mg·L^(-1),肝脏0.444 mg·L^(-1),脾脏0.351 mg·L^(-1),粘液0.276 mg·L^(-1)。脾脏、粘液和卵的提取物对3种菌均无抑菌活性,肝脏、皮肤对大肠杆菌和嗜水气单胞菌有抑菌活性,其中肝脏的抑菌活性较强。结果表明大口黑鲈体内多种组织存在着具有抗菌活性的物质。 Antimicrobial peptides were extracted from healthy largemouth bass tissues including liver,spleen,skin,mucus and ovum by 5% acetic extraction. Coomassie brilliant blue G-250 method was used to detect protein content,and bacteriostasis to Escherichia coli,Aeromona hydrophila and Staphyloccocus aureus was detected by paper-disc agar-diffusion method. The results showed that protein content of extractive part of largemouth bass was 2. 515 mg·L^-1,0. 643 mg·L^-1,0. 444 mg·L^-1,0. 351 mg·L^-1,0. 276 mg·L^-1 in egg,skin,liver,spleen,mucus. The extracts of spleen,mucus and ovum showed no antibacterial activity to 3 bacteria. The liver and skin had antibacterial activity to Escherichia coli and Aeromona hydrophila. And the liver had stronger antibacterial activity. The experiment showed that some tissues had antibacterial activity material in largemouth bass.
