摘要
为完善辣椒知识产权保护体系,补充辣椒指纹图谱库,本文通过表型差异大的8份辣椒材料筛选出的25对扩增稳定、条带清晰、有多态性的引物,建立3个簇生朝天椒杂种一代和亲本的数字化指纹图谱并进行亲缘关系分析。结果表明,对杂交种及其亲本进行扩增,共扩增出123个位点,多态性的位点51个,无多态性位点72个,多态性比率为41.5%,并构建了数字化DNA指纹图谱。分别对3个辣椒品种及其亲本进行UPGMA聚类分析结果表明,亲本间的遗传相似系数在0.36~0.59之间,母本16002-1和父本16026、16027在0.47处归为一类,父本16030单独一类。杂交种遗传相似系数在0.49~0.6之间,杂交种F16007和F16008在0.49处归为一类,杂交种F16011单独一类,与亲本的亲缘关系远近趋势一致。
In order to improve varietyintellectual property protectionsystem and supplement the fingerprint database of pepper,25 pairs of primers with stable,clear bands and polymorphic properties were obtainedsuccesfully by screening in 8 pepper varieties which showed obviously different phenotypic characteristics.Digital fingerprints were established with the 25 pairs of primers by amplifying in clustered pepper's 3 hybrids and its corresponding parents.The results showed that there were 123 loci,including 51 polymorphic loci and 72 nonpolymorphic loci,with 41.5%polymorphism rate.The digital DNA fingerprinting of three hybrids and its parents were established by using 25 pairs of InDel primers.The UPGMA cluster analysis showed that genetic similarity of 4 parents was from 0.36 to 0.59.16002-1,16026 and 16027 were clustered into one group,and 16030 belonged to the other group.The genetic similarity among hybrids was from 0.49 to 0.6.Hybrid F16007 and F16008 were clustered into one group in 0.49,and F16008 belonged to the other group.The results showed the clustered result between hybrids and parents had a relatively consistent trend.
