普鲁卡因抑制脂多糖诱导的结肠癌细胞肿瘤侵袭性的作用机制

Procaine inhibits tumorigenicity of colon cancer cells induced by lipopolysaccharide

目的探讨普鲁卡因抑制脂多糖诱导结肠癌细胞肿瘤侵袭性行为的调控及其机制。方法 CCK-8细胞活性实验检测不同浓度的普鲁卡因对结肠癌细胞活性的影响,Western blotting分析普鲁卡因对脂多糖结合蛋白表达水平的影响,划痕愈合实验检测普鲁卡因对结肠癌细胞迁移能力的影响,Transwell侵袭实验检测普鲁卡因对结肠癌细胞侵袭能力的影响。结果浓度为0.08 mg/m L的普鲁卡因对结肠癌细胞的活性影响情况最明显,差异有统计学意义(P<0.05);浓度为0.08 mg/m L的普鲁卡因对脂多糖结合蛋白的表达水平抑制最明显,差异有统计学意义(P<0.05);划痕愈合测定显示普鲁卡因可以直接抑制结肠癌细胞的运动能力;Transwell侵袭实验测定显示普鲁卡因可以直接抑制结肠癌细胞的侵袭能力,差异有统计学意义(P<0.05)。结论普鲁卡因可以通过影响脂多糖诱导结肠癌细胞的迁移和侵袭,可作为化疗辅助药物,为探索结肠癌诊疗的分子机制和新的潜在治疗策略提供相应理论帮助,为结肠癌的临床治疗方式提供有效支持。

Objective To investigate the regulation and mechanism of procaine on tumor aggressive behavior induced by lipopolysaccharide in colon cancer cells. Methods CCK-8 cell activity assay was used to detect the effect of different concentrations of procaine on the activity of colon cancer cells.Procaine lipopolysaccharide binding protein expression levels was analyzed by Western blotting.Procaine migration of colon cancer cells was detected by Scratch healing test.Effect of procaine on invasion of colon cancer cells was tested by Transwell invasion assay. Results The effect of procaine at a concentration of 0.08 mg/mL on the activity of colon cancer cells was the most significant,the difference was statistically significant（ P 〈0.05）.The concentration of procaine at the concentration of 0.08 mg/mL inhibited the expression of lipopolysaccharide binding protein most significantly,the difference was statistically significant（ P 〈0.05）.Scratch healing assays showed that procaine could directly inhibit colon cancer cell motility;Transwell invasive assay showed that procaine could directly inhibit the invasiveness of colon cancer cells,the difference was statistically significant（ P 〈0.05）.Conclusion Procaine can affect the migration and invasion of colon cancer cells by affecting lipopolysaccharide,which can be chemotherapeutic adjuvant.It provides theoretical basis for exploring molecular mechanism and new potential therapies,and provides effective supporting for treatment of colon cancer.
