The Influencing Factors in Cell Invasion and Migration Ability Testing by Transwell

对常用于检测细胞侵袭迁移能力的Transwell检测方法进行分析总结,旨在提高Transwell方法检测细胞侵袭迁移能力的一致性与稳定性。以肺癌细胞为例,对Transwell检测细胞侵袭能力中常遇到的部分细节问题进行分析,对实验中的影响因素提出应对策略,给予相应解决方案。实验发现,在Tranwell实验中,细胞种类、Transwell孔径、基质胶浓度、细胞接种数量、染色等细节问题均为该实验的关键点;并根据分析得到细胞侵袭及迁移实验中关键的影响因素,提出应对方案,可以有效提高实验结果重复性及稳定性。

In order to improve the consistency and stabilily of Transwell method, we summarized and analyzed the normalize de- tection methods of cell invasion and migration abilily by Transwell, and put tbrward the countermeasm＇es. To sum up the details of the Transwell assay in cells invasion and migration abilily by using lung cancer cells as an example, and the con＇esponding solutions were also given. The results showed that, the key points of the experiment are the lype of cell, the diameter of Transwell, the concentra- tion of matrix glue, the number of cell inoculation and the @eing, and the key influencing tactors in the experiments of cell invasion and migration are obtained, and the countermeasures are put tbrward, which can effectively improve the repeat -abilily and stabilily of the experimental results.
