摘要 针对叶绿素定量提取植物生理本科实验教学中存在的问题,该文以菠菜和油菜为实验材料,比较了研磨法与浸泡提取法两种方法,以及浸提法中不同浸提液、浸提时间对叶绿素提取效果的影响。结果显示,研磨法耗时虽然最短,但其操作琐碎复杂,容易造成叶绿素流失,结果可重复性差;叶绿素的浸泡提取法中以二甲基亚砜溶液为提取剂,操作简便,提取时间合理,且提取的叶绿素稳定,适合应用于叶绿素含量测定的实验教学。 In order to solve the problem of chlorophyll quantitative extraction in the experimental teaching of plant physiology, this paper compares spinach and rapeseed as the experimental materials, and compares the two methods of grinding and soaking exn'ac- tion, and the effect of different extraction liquid and extraction time in the extraction method on the effect of chlorophyll extraction. The results show that although the grinding method takes the shortest time, its operation is trivial and complex, which causes the loss of chlorophyll and the repeatability is poor. In the extraction of chlorophyll, two methyl sultoxide solution is used as the extractor. It is easy to operate, the extraction time is reasonable, and extracted chlorophyll is stable. It is suitable for the experiment teaching of the detennination of chlorophyll content.
