摘要
目的探讨长链非编码RNA(long non-coding RNA,LncRNA)LINC01152在乳腺癌中的作用。方法通过基因表达分析数据库(gene expression profiling interactive analysis,GEPIA)分析LINC01152在乳腺癌组织及正常组织中的表达量。根据528例LINC01152高表达的乳腺癌患者和528例LINC01152低表达的乳腺癌患者随访资料,分析LINC01152与乳腺癌患者预后的相关性,实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)法检测LINC01152在乳腺癌细胞中的表达量。生物信息学结合双荧光素酶报告基因实验探寻LINC01152潜在的转录调控因子。在BT549细胞中过表达LINC01152,在MCF-7细胞中敲低LINC01152,Transwell法检测上述细胞的侵袭能力。结果 GEPIA数据库资料显示LINC01152在乳腺癌组织中表达中位数为0.31,在正常组织中的表达中位数为2.07。生存曲线分析后发现LINC01152的表达量与乳腺癌患者预后负相关(P=0.006 5)。qRT-PCR证实LINC01152在多种乳腺癌细胞中的表达量均显著低于正常乳腺上皮细胞(P<0.01)。生物信息学预测结合双荧光素酶报告基因实验证实FOXO3a在转录水平增加LINC01152的表达。Transwell结果显示,过表达LINC01152可抑制乳腺癌细胞BT549的侵袭能力,而敲低LINC01152可明显增加MCF-7细胞的侵袭能力。结论 FOXO3a在转录水平增加LINC01152的表达,LINC01152在乳腺癌中低表达,LINC01152能显著抑制乳腺癌细胞的侵袭能力。
Objective To explore the role of lncRNA LINC01152 in breast cancer.Methods Bioinformatics analysis was performed via gene expression profiling interactive analysis(GEPIA) database to investigate LINC01152 expression in breast cancer tissues and normal tissues.The relationship between LINC01152 expression and prognosis of breast cancer patients was also analyzed by GEPIA database.Quantitative real-time PCR(qRT-PCR) assay was used to detect the expression level of LINC01152 in breast cancer cells.Bioinformatics analysis combined with dual-luciferase reporter gene assay was used to validate the potential transcriptional factors for LINC01152.The LINC01152 was overexpressed in BT549 cells and LINC01152 was silenced in MCF-7 cells,and then the cell invasion was detected by Transwell assay.Results GEPIA database suggested that LINC01152 expression was lower in breast cancer tissues than that in normal tissues,and LINC01152 was negatively correlated with poor outcome of breast cancer patients(P = 0.006 5).Meanwhile,LINC01152 expression was lower in breast cancer cells than that in normal epithelial cell by qRT-PCR assay(P 〈0.01).Bioinformatics analysis with dual-luciferase reporter gene assay showed that FOXO3 a upregulated LINC01152.Transwell assay demonstrated that LINC01152 significantly inhibited breast cancer cell invasion.Conclusion LINC01152 is reduced in breast cancer,and LINC01152 may inhibit the invasion of breast cancer cells.
