TOP2A和VEGF在宫颈鳞状细胞癌组织中的表达及其意义

Expression of TOP2A and VEGF in cervical squamous cell carcinoma tissue and its clinical significance

目的探索拓扑异构酶ⅡA(TOP2A)和血管内皮生长因子(VEGF)在宫颈鳞状细胞癌组织中的表达及临床意义。方法随机选取64例手术切除的宫颈癌组织标本和22例正常宫颈组织标本,分别采用定量PCR和免疫组织化学方法检测TOP2A和VEGF的表达情况,并分析二者表达与标本主体年龄、淋巴结转移、FIGO分期及生存情况的关系。结果 TOP2A和VEGF mRNA在宫颈鳞癌组织中的表达均明显高于正常宫颈组织(P均<0.05),并且呈正相关。在宫颈鳞状细胞癌和正常宫颈组织中TOP2A蛋白阳性表达率分别为93.8%和18.2%,VEGF蛋白阳性表达率分别为90.6%和18.2%,两者之间呈正相关(P<0.05)。TOP2A和VEGF mRNA的表达与淋巴结转移和FIGO分期相关(P均<0.05),与年龄无关(P均>0.05)。高FIGO分期和淋巴结转移阳性的宫颈鳞状细胞癌组织中TOP2A和VEGF mRNA表达显著增高(P均<0.05)。结论 TOP2A和VEGF在宫颈鳞状细胞癌中高表达,对宫颈癌的临床治疗和预后具有重要的指导意义。

Objective It is to investigate the expression and clinical significance of topoisomerase ⅡA（TOP2A） and vascular endothelial growth factor（VEGF） in cervical squamous cell carcinoma.Methods 64 cases of cervical squamous cell carcinoma tissues and 22 cases of normal cervical tissue specimens were randomly selected,in which the expression of TOP2A and VEGF were detected by quantitative PCR and immunohistochemistry respecitively,and to analyze the relationship of expression of TOP2A and VEGF with the age of the specimen,lymph node metastasis,FIGO stage and survival.Results The expression of TOP2A and VEGF in cervical squamous cell carcinoma tissues was significantly higher than that in normal cervical tissues,and showed a positive correlation between them（P0.05）.The positive expression rates of TOP2A and VEGF protein in cervical squamous cell carcinoma and normal cervical tissues were 93.8% and 18.2%,90.6% and 18.2%,respectively,and showed a positive correlation between them（P0.05）.The expression of TOP2A and VEGF in mRNA was associated with lymph node metastasis and FIGO stage（P0.05）,but not with age（P0.05）.Cervical squamous cell carcinoma tissues with high FIGO staging and lymph node metastasis had a higher level of TOP2A and VEGF mRNA expression.Conclusion TOP2A and VEGF are highly expressed in cervical squamous cell carcinoma,and have important guiding significance for clinical treatment and prognosis.
