子宫内膜异位症对大鼠卵泡发育及凋亡的影响 被引量:3

Effects of endometriosis on follicular development and apoptosis in rats

摘要 目的建立子宫内膜异位症(EM)大鼠模型,观察EM对大鼠卵泡发育及凋亡的影响。方法将33只动情周期规律的雌性SD大鼠按区组随机法分为3组,模型组(采用异体子宫移植法造模)、假手术组(开腹后相同位置丝线缝扎)、空白组(不处理),每组各10只。20天后,模型组取病灶经苏木精-伊红染色后判定模型成功率;3组均留取双侧卵巢计算卵巢指数,原位末端标记(TUNEL)染色后计算窦前卵泡、窦状卵泡及凋亡卵泡数目;取腹主动脉血,放射免疫法测定血清促卵泡生成激素(FSH)、促黄体生成激素(LH)、雌二醇(E2)、孕酮(P)水平。结果 EM模型大鼠成模率为100%。三组卵巢指数无显著差异(P>0.05)。空白组和假手术组窦前卵泡数目分别为7.65±2.40个、6.90±2.37个,窦状卵泡数目分别为13.96±2.19个、12.99±2.08个,凋亡卵泡数目分别为2.36±1.33个、2.97±1.07个,两组间无显著差异(P>0.05)。与空白组和假手术组比较,模型组窦前卵泡数目(4.79±2.08个)和窦状卵泡数目(9.42±2.26个)显著减少(P<0.05),凋亡卵泡数目(4.80±1.77个)显著增多(P<0.05);血清FSH、LH、P水平无显著差异(P>0.05),血清E2水平下降(P<0.05)。结论 EM能够促进大鼠卵泡凋亡,影响卵泡发育。 Objective A rat model of endometriosis was established to observe the effect of endometriosis(EM) on follicular development and apoptosis. Methods 33 female SD rats with normal estrous cycles were divided into 3 groups by block random,EM model group(made with allogeneic uterus transplantation),sham-operation group(open the abdomen and use silk suture the same position),the blank group(not processed). After 20 days,in the model group the lesion was stained with hematoxylin-eosin staining to determine the success rate of the model. In all 3 groups,ovaries were retained to calculate ovarian index. The apoptotic follicles was investigated using terminal deoxynucleotidyl transferase-mediated d UTP nick end labeling technique,and the number of preantral,antral and apoptotic follicles was calculated. Abdominal aortic blood was collected,and the level of serum FSH,LH,E2 and P were determined by radioimmunoassay. Results The success rate of endometriosis model rats was 100%. There was no significant difference in ovarian index between the three groups(P〉0. 05). In the blank group and sham-operation group the number of preantral follicles was 7. 65 ± 2. 40 and 6. 90 ± 2. 37,the number of antral follicles 13. 96 ± 2. 19 and 12. 99 ± 2. 08,the number of apoptotic follicles 2. 36 ± 1. 33 and 2. 97 ± 1. 07,and there was no significant difference between the two groups(P〉0. 05).Compared with blank group and sham-operation group,in the model group rats,the number of preantral and antral follicles(4. 79 ± 2. 08,9. 42± 2. 26) decreased(P〈0. 05),while the number of apoptotic follicles(4. 80 ± 1. 77) increased(P〈0. 05),the level of serum FSH,LH and P showed no significant difference(P〈0. 05),but the level of serum E2 decreased(P〈0. 05). Conclusion EM increases follicular apoptosis and affects follicular development.
