摘要
为了解2016年山东地区H_9N_2亚型禽流感病毒的变异情况,选取2016年所分离的10个H9N2亚型禽流感病毒株,对其HA基因进行测序,并应用DNAStar和Mega5.0生物软件拼接序列和建立进化树。结果表明,10株H_9N_2分离毒的HA裂解位点氨基酸均为-RSSR/GLF-,有6~8个潜在的糖基化位点,具有典型低致病性禽流感病毒HA基因的特征,但其226位氨基酸均为亮氨酸(L),呈现出人流感受体结合特性。进化树分析表明,2016年山东地区H_9N_2流行毒株HA基因属于A/Duck/Hong-Kong/Y280/97亚群,同属于近几年在中国鸡群中流行的以A/chicken/Zhejiang/HJ/2007为代表的G57分支。
To understand the variation of H9N2 subtype avian influenza virus (AIV)in Shandong in 2016, 10 strains of H9N2 subtype AIV in 2016 were selected and their HA gene was sequenced and analysed by software DNAStar and Mega5.0.The results showed that the amino acids in the HA cleavage sites were all -RSSR/GLF-, with 6-8 potential glycosylation sites. The HA genes had the typical low pathogenicity avian influenza virus features,but its 226 amino acids were leucine (L), showing the binding characteristics of human influenza receptors. Evolutionary tree analysis showed that HA genes of H9N2 epidemic strain from Shandong in 2016 belonged to A/Duck/Hong-Kong/Y280/97 subgroup, and co-belonged to the G57 branch represented by A/chicken/Zhejiang/HJ/2007, which was prevalent in chickens in China in recent years.
