摘要
目的:以金银花连翘药中的连翘酯苷A、木犀草苷、连翘苷为主要研究对象,设计动物实验方案,以确定金银花连翘药对在发热SD大鼠体内各组织中有效成分的分布规律。方法:正常大鼠7只,除空白组外每只给药5 m L,1 h、2h、3 h、5 h、7 h、9 h后分别取心、肝、脾、肺、肾,匀浆,涡旋后离心,取上清液使用HPLC法测定各组织药物含量。结果:结果显示连翘酯苷A在肝脏中5 h都达最大值且含量最多。木犀草苷在心中2 h达最大值,7 h在心、肾中,9 h在肺中时含量也较大,其他组织较少。连翘苷在肾中2 h时含量达最大值,在其他组织中含量相差不大。结论:灌胃后,药物在体内迅速分布,经过肝脏吸收后重新分配到全身各处。心和肺中有效成分含量较低,进一步用科学理论证明了金银花连翘皆"轻清宣散,偏于清透上半身之邪",为金银花连翘药对的作用特点提供了科学理论依据。
Objective:In this paper,forsythoside A,cynaroside and phillyrin are the main study objects. In order to determine the distribution of the effective components of the drug pairs of Flos Lonicerae and Fructus Forsythiae,which distribute in the fever SD rats,it designs experiments on animals. Methods: Except the blank group,every 5 m L drug was infused to stomach of each mouse. After 1 h、2 h、3 h、5 h、7 h、9 h,organs were taken out. First,we homogenized it separately. After vortexing it,we used centrifugalization. Finally,we got supernatant to analyze the pharmic content by HPLC. Results:The results showed that forsythoside A reached the maximum and the content also reached the maximum in liver in 5 h. When in 7 h,the cynaroside reached the larger value in heart and kidney,and it also reached the larger value in lung in 9 h than that in other organs. But it reached the maximum in heart in 2 h. The phillyrin reached the maximum in kidney,however,the content in other organs were almost the same. Conclusion:After filling the stomach,the drug is rapidly distributed throughout the body and redistributed after being absorbed by the liver. The phenomenon of low effective component content in the heart and lungs,further use scientific theories to prove the honeysuckle forsythia are‘clearing away the heat-evil to relieve the exterior with light drug',to provide scientific theoretical basis for characteristic of the drug pairs of Flos Lonicerae and Fructus Forsythiae.
