多指标优选糖网黄斑水肿方颗粒提取工艺及蒲黄影响溶出的机制考察 被引量:1

摘要 目的:优选糖网黄斑水肿方颗粒提取工艺,并考察蒲黄影响黄芪药效成分溶出的机制。方法:采用HPLCELSD法测定黄芪甲苷含量,HPLC-DAD法同时测定毛蕊异黄酮葡萄糖苷、香蒲新苷、异鼠李素-3-O-新橙皮苷含量。以4个成分为评价指标,优选蒲黄提取方式。以蒲黄指标成分为评价指标,单因素法优选蒲黄提取工艺。以黄芪指标成分和干膏率为评价指标,以综合评分法建立三因素三水平正交试验,优选黄芪等7味药提取工艺。通过测定不同阶段下黄芪指标成分的变化,考察蒲黄对黄芪成分溶出的影响。结果:最佳提取工艺为:称取蒲黄,加11倍水,煎煮4次,每次50 min,过滤,得蒲黄单煎液;称取黄芪等7味药,加8倍水,煎煮3次,每次40 min,过滤,得黄芪等7味药合煎液。蒲黄的吸附作用是影响黄芪成分溶出的主要原因。结论:糖网黄斑水肿方颗粒中蒲黄粉末和黏液质具有吸附作用,影响黄芪指标成分溶出,制备时蒲黄应采用单煎法。优选的工艺合理可行、稳定科学,适合于该制剂的工业化生产。 Objective:To optimize the extraction technology of Tangwanghuangban Shuizhongfang Granule,and investigate the dissolution mechanism of active constituent of Astragali influence by Pollen Typhae. Methods:HPLC-ELSD was employed to determine content of astragaloside Ⅳ,HPLC-DAD was employed to simultaneous determine content of calycosin glycoside,typhaneoside,isorhamnetin-3-O-neohesperidoside. Using the 4 constituents as the evaluate index,optimize the best extraction type of Pollen Typhae. Using the index constituents of Pollen Typhae as the evaluate index,optimize the best extraction technology of Pollen Typhae by single factor test. Using the indexes constituents and yield of dry extract as the evaluate index,optimize the best extraction technology of other 7 herbs such as Astragali by orthogonal test. By measuring the change of index constituents of Astragali in different extract stages,investigated the dissolution mechanism of active constituents of Astragali influence by Pollen Typhae. Results:The best extraction technology was as follows:Pollen Typhae,extracted 4 times with 10 times amount of water,50 mins for per time,filter fluid as the single decoction of Pollen Typhae;other 7 herbs such as Astragali,extracted 3 times with 8 times amount of water,40 mins for per time,filter fluid as the combined decoction of 7 other herbs. The adsorption effect of Pollen Typhae is the main factor that influence the dissolution of active constituents of Astragali constituents. Conclusion:Pollen Typhae,as a herb of Tangwanghuangban Shuizhongfang Granule,with feature of powder and mucilage,which is the main reason that influence the dissolution of active constituents of Astragali,so single extraction technology should be adopted in Pollen Typhae. The optimized technology is reasonable and feasible,stable and scientific,and it can provide guidance for industrial production.
