摘要
目的:观察脐疗对寒凝血瘀型痛经模型大鼠子宫肥大细胞数目及脱颗粒率的影响,部分阐释其效应机制。方法:将50只雌性Wistar大鼠随机分为5组:空白组、模型组、隔药灸脐组、药物敷贴组、艾条灸脐组。除空白组外均构建寒凝血瘀型痛经模型,各治疗组分别给予相应治疗。采用改良甲苯胺蓝染色法、免疫荧光技术,检测各组大鼠子宫肥大细胞数目及脱颗粒率。结果:寒凝血瘀型痛经模型组大鼠子宫肥大细胞数目及脱颗粒率均高于空白组大鼠(P〈0.01)。治疗后子宫肥大细胞数目:隔药灸脐组、艾条灸脐组均低于模型组(P〈0.01),隔药灸脐组、艾条灸脐组均低于药物敷贴组(P〈0.01),隔药灸脐组低于艾条灸脐组(P〈0.05)。治疗后子宫肥大细胞脱颗粒率:隔药灸脐组低于模型组(P〈0.01),艾条灸脐组低于模型组(P〈0.05),隔药灸脐组低于药物敷贴组(P〈0.01)。结论:隔药灸脐法、艾条灸脐法均能通过抑制寒凝血瘀型痛经大鼠子宫肥大细胞的活化而达到治疗目的,其中隔药灸脐法疗效最明显。
Objective:By constructing the model of cold coagulation and blood stasis type of dysmenorrhea,the effect mechanism of umbilical therapy on the treatment of cold coagulation and blood stasis type of dysmenorrhea was discussed from the angle of mast cell. Methods:Rats were randomly divided into 5 groups:normal group( group A),model group( group B),the group of herb-partition moxibustion on the navel( group C),the group of drug covering navel therapy( group D) and the group of navel therapy with moxa stick moxibustion( group E). Rats in the umbilical therapy groups and the model group were constructed as cold coagulation and blood stasis type of primary dysmenorrhea model. Each group was given the corresponding treatment from the sixth day of the start of modeling,once a day,for 5 times in total. After treatment and extraction of tissue,using modified toluidine blue staining,immunofluorescence methods were used to test the mast cells number in rats' uterine. Results:The numbers of mast cells in uterine tissue of group C and group E were all less than those of the group B( P〈0. 01) and group D( P〈0. 01). The number of mast cells of group C was less than that of the group E( P〈0. 05). The degranulation rate of mast cells in uterine tissue of group C was less than that of group B( P〈0. 01). The degranulation rate of mast cells of group E was less than tha tof group B( P〈0. 05). The degranulation rate of mast cells of group C was less than that of group D( P〈0. 01). Conclusion:The herb-partition moxibustion on the navel and navel therapy with moxa stick moxibustion could inhibit the activation of mast cells in uterine tissue of cold coagulation and blood stasis type of dysmenorrhea model rats. The therapeutic advantages of herb-partition moxibustion on the navel are more obvious than the two other groups. The difference of curative effect may be reflected in the difference of Ca2 +activation.
