摘要
目的建立QuEChERS-高效液相色谱-串联质谱法测定火锅底料中5种罂粟壳生物碱的分析方法。方法火锅底料用QuEChERS方法去除杂质,选择Agilent Eclipse Plus C18色谱柱(100 mm×4.6 mm,3.5μm),以0.01 mol/L乙酸铵溶液(含0.1%甲酸)和乙腈作为流动相进行梯度洗脱,在0.4 m L/min流速下,以多反应监测(multiple reaction monitoring,MRM)扫描模式进行检测,外标法定量。结果吗啡、可待因、蒂巴因、罂粟碱、那可丁5种罂粟壳生物碱在火锅底料中的回收率范围为50.3%~105.4%,精密度范围为1.79%~7.91%,方法检出限分别为1.25、1.25、0.25、0.25、0.25μg/kg。结论该方法简便、快速、可靠,可用于火锅底料等食品中罂粟壳生物碱的快速筛查与确证。
Objective To establish a method for the determination of 5 kinds of pericarpium papaveris alkaloids in hot pot seasoning by QuEChERS-high performance liquid chromatography-tandem mass spectrometry. Methods Hot pot seasoning was purified by QuEChERS. The liquid was performed on Agilent Eclipse Plus C_(18)(100 mm×4.6 mm, 3.5 μm) with gradient elution of ammonium acetate solution(containing 0.1% formic acid) and acetonitrile, and the flow rate was 0.4 m L/min. The sample was detected in multiple reaction monitoring(MRM) scan mode, and quantified by external standard method. Results The recoveries rate of morphine, codeine, tibaine, papaverine and nacodin at 3 additional levels were 50.3%-105.4%, the relative standard deviations were 1.79%-7.91%, and the limits of detection of the method were 1.25, 1.25, 0.25, 0.25, 0.25 μg/kg. Conclusion This method is simple, fast and reliable, which can be used for rapid screening and confirmation of poppy shell alkaloids in foods such as hot pot seasoning.
