摘要
目的:探讨摄入不同剂量某54%vol白酒对大鼠肾上腺髓质细胞内肾上腺髓质素(ADM)及促肾上腺皮质激素(ACTH)表达的影响。方法:正常雄性SD大鼠120只随机法分为正常对照组(NG,n=30)和实验组(灌胃白酒,n=90),实验组又分为低剂量组[LG,0.8 m L/(kg·d)]、中剂量组[MG,1.6 m L/(kg·d)]和高剂量组[HG,2.4 m L/(kg·d)],实验组大鼠于每日11∶00 AM及5∶00 PM各灌胃一次,NG组不予处理;各组大鼠分别于实验第4、8、12、16及20周末取肾上腺组织,采用免疫组织化学SABC法、图像分析法、Western blot检测大鼠肾上腺髓质中ADM及ACTH表达。结果:与NG相比较,第4、8、12、16及20周末各实验组,大鼠肾上腺髓质细胞ADM、ACTH免疫组织化学染色强度及平均光密度差异均无统计学意义(P>0.05);Western blot显示,与NG组比较,第4、8、12、16及20周各实验组大鼠肾上腺组织ADM、ACTH蛋白表达水平差异均无统计学意义(P>0.05)。结论:在本实验设定的剂量和时长下,摄入该54%vol白酒对大鼠肾上腺髓质ADM及ACTH的表达无明显影响。
Objective: To investigate the effects of different doses of certain 54% vol Liquor on the expression of adrenomedullin( ADM) and adrenocorticotropic hormone( ACTH) in rat adrenal medulla cells. Methods: 120 normal male SD rats were randomly divided into normal control group( NG,n= 30) and experimental group( EG,n = 90). The experimental group was divided into low dose group[LG,0. 8 mL/( kg·d) ],middle dose group [MG,1. 6 mL/( kg·d) ]and high-dose group [HG,2. 4 mL/( kg·d) ]. Rats in the experimental group were given intragastric administration at 11∶ 00 a.m. and 5∶ 00 p. m. daily,but no treatment was given in the normal group. The adrenal tissues of each group were collected at the end of the 4 th,8 th,12 th,16 th and 20 th week. The expression of ADM and ACTH in the adrenal medulla was detected by immunohistochemical SABC method,image analysis method and Western blot. Results: The immunoreactive products of ADM and ACTH in normal and experimental groups were brown or claybank,and distributed in the adrenal medulla cytoplasm/karyon. At the end of the 4 th,8 th,12 th,16 th and 20 th week.,there was no significant difference in immunohistochemical staining intensity and average optical density of adrenal medulla cells between the experimental group and the normal control group( P〉0. 05). Western blot showed that there was no significant difference in the expression of ACTH protein in adrenal tissue of rats in the 4 th,8 th,12 th,16 th and 20 th week compared with the normal control group( P〉0. 05). Conclusion: Under the dosage and duration of the experiment,the expression of ADM and ACTH in the adrenal medulla of rats was not significantly affected by the intake of the 54% vol liquor.
