人食管鳞癌细胞球细胞的干细胞特性研究 被引量:3

摘要 目的探讨不同代次食管癌Eca109细胞球细胞p75^(NTR)核阳性表达情况和增殖、侵袭能力。方法运用无血清成球培养法,富集到第1代的肿瘤干细胞球样细胞,对其进行离心、接种,得到第2代直至第9代细胞球细胞。检测第1代、3代、5代、7代、9代细胞球细胞中肿瘤干细胞特异性标志物Ki67和p75^(NTR)的表达情况,并对细胞增殖、侵袭能力进行对比。结果第1代、3代、5代细胞球细胞中p75^(NTR)核阳性表达的细胞数量随球细胞代数的增长依次递增,增殖、侵袭能力都随之相应增强;第5代、7代、9代细胞球细胞核均p75^(NTR)表达阳性,同时其相应的增殖、侵袭能力均未见显著差异。结论食管癌p75^(NTR)核阳性表达的球细胞可能是食管癌干细胞。 Objective To investigate the expression of p75^NTR in nuclear, the proliferation and invasion of cell sphere derived from the esophageal cancer cell line Eca109. Methods The Eca109 cell line was cultured in serum-free medium to enrich the first generation of tumor-derived stem cells, and then cells were centrifuged and cultured again to get the second generation sphere cells,,until to get the 9 th generation sphere cells, respectively. The expression of Ki67, p75^NTR , the cell proliferation and invasion were detected in different generation sphere cells. R esults The number of cells that p75^NTR expressed in nuclear increased along with the generations of the cell sphere from the first generation to the 5 th generation cell spheres, and the spheres formation, proliferation and invasion was increased accordingly, but from the 5 th generation to the 9 th generation cell spheres, the sphere cells were all the p75^NTR positive in nuclear and the spheres formation, proliferation and invasion was not significantly different along with the cell spheres generations. Conclusion It can be concluded that the cells that p75^NTR expressed in nuclear of the spheres cells may be esophageal cancer stem cells.
