大孔吸附树脂纯化紫穗槐果实总黄酮的研究 被引量:3

摘要 本研究采用D-101大孔吸附树脂分离纯化紫穗槐果实中的总黄酮,研究了影响树脂静态和动态吸附与洗脱的主要因素,以单因素为基础,研究上样液浓度、上样液流速、洗脱剂的体积分数、洗脱剂流速对总黄酮分离纯化的影响,并找到最佳工艺条件。结果表明,D-101型大孔吸附树脂对紫穗槐果实中总黄酮静态吸附和解吸附最佳工艺条件为:上样液浓度0.86mg/mL,吸附平衡时间为6.5h,洗脱剂乙醇体积分数80%,解吸附平衡时间为1.8h。其对紫穗槐总黄酮动态吸附和洗脱最佳工艺条件为:准确称取大孔吸附树脂15g装柱,床体积(BV)约30mL。以体积为30mL、浓度为0.75mg/mL的紫穗槐总黄酮粗提液过柱,流速为1.0mL/min,充分吸附后,再用3BV的超纯水洗柱,最后用50mL 80%乙醇溶液以流速1.5mL/min进行洗脱。在此工艺条件下,能有效地洗脱色素、叶绿素等非目标成分,科学合理的分离纯化紫穗槐果实总黄酮,且操作简单、安全、成本低廉。 In this study,D-101 macroporous adsorption resin was used to separate and purify total flavonoids from fruits of Amorphafruticosaand studied the main factors affecting the static and dynamic adsorption and elution of resin.Based on a single factor,the influence of the concentration of the sample liquid,the flow rate of the sample solution,the volume fraction of the eluent and the flow rate of the eluent on the separation and purification of the total flavonoids were studied to find the best process conditions.The results showed that the optimum conditions of static adsorption and desorption of D-101 macroporous resin for total flavonoids of Amorpha fruticosa were as follows:the concentration of the upper sample was0.86 mg/mL,the adsorption equilibrium time was 6.5 h,the ethanol volume fraction of the eluent was 80%,and the desorption equilibrium time was 1.8 h.The optimum conditions for the dynamic adsorption and elution of total flavonoids of Amorpha fruticosa were as follows:the macroporous resin 15 g was packed accurately and the bed volume(BV)was about 30 mL.The crude extract of Amorpha fruticosa with the volume of 30 mL and the concentration of0.75 mg/mL was over the column,the flow rate was 1.0 mL/min,after adsorbed fully,then the column was washed with 3 BV(bed volume)ultrapure water,and finally eluted with 50 mL of 80% ethanol solution with a flow rate of 1.5 mL/min.Under this process conditions the non-target ingredients such as pigment and chlorophyll can be eluted effectively,and the total flavonoids of the fruit of Amorphafruticosaare separated and purified scientifically and reasonably,and the operation is simple,safe and low cost.
