摘要
目的:研究乙肝癌变肝组织中HBX、hTERT的表达水平及意义。方法:以动物实验研究HBx对肝肿瘤形成与转移的影响,并以TRAP-PCR-ELISA、DNA印迹法及RT-PCR明确HBx对hTERT产生的直接调控作用。结果:HBx稳定表达裸鼠动物模型肿瘤体积、肿瘤重量均明显大于表达敲低裸鼠动物模型(P<0.05),且产生转移,而HBX表达敲低裸鼠动物模型无转移;HBx稳定表达裸鼠动物模型细胞有明显梯状条带,而HBx表达敲低裸鼠动物模型细胞则无明显梯状条带;HBx稳定表达裸鼠动物模型端粒长度(4.28±1.06)kb,明显短于HBx表达敲低裸鼠动物模型(9.64±3.67)kb(P<0.05),且细胞hTERT m RNA表达量为(76 028.00±13 685.12),明显高于HBx表达敲低裸鼠动物模型(42 536.00±8 963.24)(P<0.05)。结论:HBx能够通过各类途径产生致癌作用,其中包括激活hTERT,导致细胞出现永生化癌变,故HBX、hTERT表达水平的检测可为乙肝癌变临床诊治提供指导。
Objective:To study the expression of HBX and hTERT in hepatitis B cancerous liver tissues and its significance.Methods:The effect of HBx on the formation and metastasis of liver tumors was studied by animal experiments.The direct regulation effect of HBx on hTERT was determined by TRAP-PCR-ELISA,DNA blotting and RT-PCR.Results:The tumor volume and tumor weight of nude mice with stably expression of HBx were significantly higher than those of the nude mice with expression knock-down of HBx(P〈0.05).There was metastasis in the former while there was no metastasis in the latter.Cells of the former mice showed ladder-like band while cells of the latter showed none ladder-like band.The telomere length of the former was significantly shorter than that of the latter [(4.28±1.06) kb vs(9.64±3.67) kb(P〈0.05) ],and the expression quantity of hTERT m RNA in cells was significantly higher than that in HBx knock-down nude mice models [(76 028.00±13 685.12) vs(42 536.00±8 963.24)(P〈0.05) ].Conclusion:HBx can produce carcinogenic effects by various methods,including activation of hTERT,resulting in cell immortalization and carcinogenesis.Therefore,the detection of the expression levels of HBX and hTERT can provide guidance for clinical diagnosis and treatment of hepatitis B canceration.
