摘要 目的探讨PM2.5对小鼠过敏性哮喘模型炎症反应的影响。方法 27只BALB/c雌性小鼠随机分成对照组、卵清蛋白(OVA)组、OVA+PM2.5组;OVA组、OVA+PM2.5组于第1、8、15天用OVA腹腔注射致敏,并从第16天起每天用OVA雾化吸入激发1次共7次,其中OVA+PM2.5组在第16天气管内滴注PM2.5处理;对照组用生理盐水处理。分别在第17、20、23天评估支气管反应性,检测肺泡灌洗液中白细胞分类及炎症性细胞因子水平。结果与对照组、OVA组比较,OVA+PM2.5组支气管肺泡灌洗液中嗜酸性粒细胞、中性粒细胞、巨噬细胞数量及IL-4水平、支气管反应性升高,而IFN-γ水平下降(P<0.01或0.05)。结论 PM2.5加重哮喘小鼠支气管反应性及炎症反应。 Objective To investigate the effect of PM2.5 on inflammatory response in a murine model of allergen-induced asthma. Methods Twenty-seven female BALB/c mice were randomized to control, ovalbumin(OVA), and OVA+PM2.5 groups. OVA and OVA+PM2.5 groups were sensitized with OVA at days 1, 8 and 15 and rechallenged with OVA once daily at day 16 for 7 days, of which OVA+PM2.5 group were simultaneously rechallenged with PM2.5 at day 16. Control group were treated with normal saline. Bronchial reactivity and leukocyte differential count and inflammatory cytokines in bronchoalveolar lavage fluid(BALF) were determined at days 17, 20 and 23. Results Compared with control and OVA groups, levels eosinophils, neutrophils, macrophages and IL-4 in BALF and bronchial responsiveness were higher but IFN-γ content was lower in OVA+PM2.5 group(P〈0.01 or 0.05). Conclusion PM2.5 can exacerbate the bronchial responsiveness and inflammation in allergic asthma mice.
