miR-381通过调控NF-κB影响膀胱癌细胞增殖及凋亡 被引量:8

miR-381 influences proliferation and apoptosis of bladder cancer cell through regulating NF-κB

摘要 目的了解mi R-381对膀胱癌UM-UC-3细胞增殖及凋亡的影响。方法 mi R-381 mimic和pc DNA NF-κB转染UMUC-3细胞,CCK8法检测细胞增殖,流式细胞术检测细胞凋亡,Western blot检测NF-κB和Bcl-2表达。结果 mi R-381 mimic抑制UM-UC-3细胞增殖,促进细胞凋亡,减少NF-κB和Bcl-2表达;pc DNA NF-κB阻滞mi R-381 mimic对UMUC-3细胞增殖和凋亡的作用。结论 mi R-381可能通过调控NF-κB抑制膀胱癌细胞增殖、诱导细胞凋亡。 Objective To investigate the effect of mi R-381 on proliferation and apoptosis of bladder cancer UM-UC-3 cells.UM-UC-3 cells were transfected with mi R-381 mimic and pc MethodsDNA NF-κB.The proliferation,apoptosis,and expression of NF-κB and Bcl-2 were determined by CCK8 assay,flow cytometry and Western blot,respectively.ResultsMi R-381 mimic suppressed proliferation and levels of NF-κB and Bcl-2,but enhanced apoptosis in UM-UC-3 cells.However,these effects were blocked by pc DNA NF-κB.Conclusion mi R-381 may inhibit proliferation and induce apoptosis via regulating NF-κB in bladder cancer cells.
